Objective To investigate the effects and mechanisms of Yiyanling granule on experimental severe acute pancreatitis (SAP) in Rats. Methods 125 rats were randomized into 6 groups: sham operation group (SO,n=20), SAP group (n=25),SAP and yitaimei group (SAP rats were treated with Yitaimei n=20), SAP and 3 Yiyanling granule dose groups (SAP rats were treated with big, middle and small dose Yiyanling granule n=20 /group). The SAP model was induced by bili-pancreatic duct infusion with 5% sterile sodium taurocholate solution. Tumor necrosis factor(TNF) , interleukin-6(IL-6) , interleukin-8(IL-8) were determined by using radioimmunoassay, serum malondialdehyde (MDA) was determined by using thiobarbiturate, amylase in serum and urine were determined by the blue-starch method, blood serum lipase was determined by using turbidimetric and Lipopolysaccharide(LPS) was measured by using limulus test, etc. Results The amylase in serum and urine , lipase in blood serum , MDA level of blood plasma, TNF, IL-6, IL-8, and LPS in rats with SAP were increased and the amount of them were greatly decreased in yitaimei and 3 Yiyanling granule dose groups (P<0.05) . Conclusion Yiyanling granule can inhibit the pancreatic enzymes activity, correct the imbalance of oxygen free radical metabolism, eliminate LPS in serum, and adjust metabolism of cytokines. Yiyanling granule has an obvious healing effect on severe acute pancreatitis in rats and plays a positive role on stabilizing and controlling symptoms.
