Objective: In order to seek an central mechanism of WPD capsule that cures PMS with liver-qi invasion rat, we have observed if the model rat drug serum has an effect on the primary cultured nerve energy and serotonin 1A receptor of rat pallium. Methods: First paeoniflorin level of the model rat serum was detected by HPLC in different time after the treatment of WPD capsule. According to the peak time of paeoniflorin concentration-time change curve, we got the rat blood for drug serum. Second we made use of Giemsa colouration and immuno-fluorescent-chemistry to observe and identify the cultured nerve respectively. Third we studied the drug serum effect of PMS with liver-qi invasion rat on primary cultured nerve energy and serotonin 1A receptor of rat pallium using the MTT and immuno-fluorescent-chemistry respectively. Results: Compared with the model group,both rat pallium nerve MTT value and its serotonin 1A receptor in high dose group increased markedly(P<0.01), nerve MTT value and its serotonin 1A receptor in middle dose group aslo enhanced (P<0.05, P<0.01), no significant changes were observed in low dose group(P>0.05). In comparison with the normal group.the serum of the model group decreased the nerve MTT value observably (P<0.05), but serotonin 1A receptor decreased slightly(P>0.05). The change of nerve MTT value and its serotonin 1A receptor in the drug serum groups presented a dose-effect relation. Conclusion: WPD capsule can rectify the decrease trend of PMS liver-qi invasion rat nerve serotonin 1A receptor level. Maybe it is just the central mechanism of the new drug curing this syndrome.
