Objective The experiment was based on the findings of pervious animals study. We made the PMS liver-qi depression rat model with the methods of multiple factors including emotional stimulation.Our aim is to investigate the effect of Jingqianshu granule on transcriptions of ERp and related gene 5-HT1AR,5-HTT,MAO-A/B in primary rat hippocampus neurons, as well as to study the pathogenesis of PMS liver-qi depression to provide reliable experimental data for the clinical treatment of PMS liver-qi depression.Methods We confirmed the menstrual pahases of rats by observing the vagina cell shape and selected the rats at the non-acceptance period,then randomly divided the rats into three groups, including normal control group, PMS liver-qi depression model group and Jingqianshu granule treatment group. Serum of rats in three groups were collected and then added into the medium of in vitro cultured hippocampi primary neurons. At last the mRNA levels of the ERβand related gene 5-HT1AR,SERT,MAO-A/B were detected with RT-PCR respectively.Results Bondaging the limbs of rats could stimulate the process of clinical emotional abnormalities that triggered PMS liver-qi depression successfully. Compared with the normal control group,the serum of rats in model group obviously improved mRNA levels of the ERβand MAO-A (P<0.05),and markedly decreased mRNA levels of the 5-HT1AR and 5-HTT (P<0.05).While serum of rats in Jingqianshu granule improve the abnormal conditions of mRNA levels significantly (P<0.05), which had no difference with the control group. The MAOB mRNA levels of three groups had no statistical difference (P>0.05)Conclusion The method of bondaging the limbs of rats is successful. There is a intimate relationship between the mRNA levels of the ERβ,5-HT1AR,5-HTT and MAO-A with PMS liver-qi depression, which maybe one of the mechanism treating PMS liver-qi depression. But the MAOB had little relevance with liver-qi depression.
