OBJECTIVE To approach the protection and mechanism of action of Anisodamine(654 -2) on transplantation of pancreas in rats. METHODS Establish the model of transplantation of pancreas in rats. 210 Wistar rats were divided into 4 groups randomly, with 30 in SO ( sham operation) group, and 30 pairs in each of the other 3 groups. SO group : treated only with abdominal cutting ; Control group: made to be model of transplantation of pancreas; Experimental group A; treat each recipient with Anisodamine 5mg/kg i. v. ,10 min before reperfusion in transplantation of pancreas, with anything else the same as Control group; Experimental group B;treat each recipient with Anisodamine 10mg/kg i. v. ,10 min before reperfusion in transplantation of pancreas, with anything else the same as Control group. Except in SO group, in other groups blood was drawn and pancreatic tissue was cut at 0,2,4,8,12h after reperfusion,and the levels of blood amylase,TXB2, 6 - K(6 - keto - prostaglandin F1α ) were determined, and change of histopathology of transplanted pancreases were observed. RESULTS: 2h later,blood amylase, TXB2 and 6 - K were all up;4h later, TXB2/6 - K ratio was in disturbance, and pancreatic patho - injury was obvious. Anisodamine can reduce levels of TXB2 and blood amylase (P <0.05) and patho - injury,and relieve disturbance of TXB2 /6 - K ratio. CONCLUSIONS Anisodamine have obvious protection to transplantation of pancreas in rats, and its mechanism of action is related to inhibition of TXB2, relief of disturbance of TXB2/6 - K ratio, improvement of microcirculation and relief of ischemical reperfusion injury.
