objective: Heat shock protein ( HSP ) is also called molecular chaperones that all cells can produce under stress conditions, Hsp70 is a part of the HSP family which contains a number of highly-related protein isoforms ranging in size from 66kDa to 78kDa,Including HSP20,HSP70 ,HSP90 and macromolecular family.HSP70 is one of the most important member in HSP family. HSP70 is closely related to biological functions. The molecular chaperones of the Hsp70 family is recognized bind to nascent polypeptide chains as well as partially folded intermediates of proteins preventing their aggregration and misfolding. Members of the Hsp70 family are molecular chaperones which are involved in many cellular functions such as protein folding, transport, maturation and degradation. At present the study of HSP has become hotspot, but their specific functions havn't been recognized yet . Especiaclly the study about the influence of medium in the formation of HSP has not been reported. The objective of this test is to explore the influence factors through expression of HSP70 on peripheral blood mononuclear cells under various pH serum and different medium . Method:Whole blood was taken from 16 healthy volunteers without fever and inflammation , PBMC were separated by Percoll density gradient centrifugation., then hot shocked in different pH serum and various medium including 1640 media , type AB serum and phosphate buffer solution (PBS) under 42℃water bath,0.5 and 1 hour , HSP70 was detected by immuno histochemical staining Results:Compare with the blank, the expresson of HSP70 on PBMC hot shocked under 42℃,0.5 and 1 hour in different pH serum were significantly difference, P<0.01. The expressions of HSP70 on PBMC in type AB serum(pH7.35)hot shocked under 42℃ for 0.5 and 1 hour, were significantly difference, P<0.01, the expressions of HSP70 within 1 hour is greater than that of 0.5 hour; The expressions of HSP70 PBMC under acide condition of type AB serum(pH6.95)hot shocked for 0.5 and 1 hour, there was no statistically significant difference,P>0.05; The expressions of HSP70 on PBMC under alkaosis type AB serum(pH7.65)hot shocked 0.5 and 1 hour, there was no statistically significant difference , P>0.05. in 1640 medium was significantly different compared with that in serum P<0.01 .There was no statistically significant difference between 0.5 and 1hour in 1640 medium The expressions of HSP70 on PBMC hot shocked under 42℃in various media were significantly different as compared with that of blank ,P<0.01. PBMC hot shocked under 42℃for 0.5 hour in PBS, the expression of HSP70 was significantly different compared with that of 1 hour,P<0.01, the expressions of HSP70 within 0.5 hour is greater than that of 1 hour; PBMC hot shocked under 42℃for 0.5 ,1 hour in 1640 medium, the expressions of HSP70 there was no statistically significant difference , P>0.05. Conclusion: (1)The expressions of HSP70 on peripheral blood mononuclear cells is related to the number of stress source closely. multiple stresses result in fast formation of HSP70 than single stress does. (2)Cells can produce hot shock protein under alkalosis condition. (3) The expressions of HSP70 on peripheral blood mononuclear cells is related to the media.
