IntroductionPremature rupture of fetal membrane ( PROM) is composed of two types, one is term premature rupture of fetal membrane ( TPROM) , and the other is preterm premature rupture of fetal membrane ( PPROM ). The incidence of TPROM is about 10 percent,and that of PPROM is about 2 to 3.5 percent.As we all know, many serious complications always follow PROM, such as intraamniotic infection ( Chorioamnionitis in parthology) , premature delivery, prolapse of cord, placental abruption amniotil fluid embolism, asphyxia of fetus and etc. All these complications above will inevitably increase the incidence rate and death rate of pregnancy women and their fetus. But up to now, Many scholars are paying more attention to prom and investigations of the mechanisms of PROM are still going on. From this study, NF -кB may be one of the factors which participate in the happening of PROM. IL - 15 is a kind of new Th1 -type cytokines , and it has many biological Junctions like IL -2. NF - кB can induce the IL - 15 expression, and IL - 15 can stimulate NF - кB activation. We also studied the percentage of NF - кB P65 positive cells in the tissue of fetal membrane and the IL - 15 levels in the blood of mother in order to find their roles in PROM.Materials and Methods— The source of materials:I selected thirty PROM patients as examine cases. They all delivered at the second Affiliated Hospital of China Medical University from March,2004 to December ,2004. These cases are all at the age of 23 to 39, average 28. 67 ±4. 03, and the gestation weeks from 37 to 39+4,average 38. 31 ±0.70. These cases didn't use antibiotic drugs before, and hadnt any clinical infection signs or other complications during pregnancy except PROM. They were divided into twogroups according to the lasting time of membrane rupture. One group: the lasting time of membrane rupture was not longer than 24 hours; the other group: the lasting time of membrane rupture had been more than 24 hours.Using the pathological standards, these cases can also be divided into two groups: chorioamnionitis group and non - chorioaminionitis group. In this stud-y, I setect thirty normal pregnancy women acting as the control cases. These cases are all at the age of 23 -39. average 29.57 ±3.42 and the gestation weeks from 37 - 39 +6, average 38.62 ±0.46. Compared with PROM cases, they have no obvious diffrence in age and gestation week (P >0.05).ReagentsNF - кB P65 monoclonal antibodyIL —15 ELISA reagent boxMethods1, collection of the casesI collected 2ml blood in elbow veins of all pregnancy women. The blood was centrifuged at 1000 rpm for 15 minutes,, and then stored at -20℃. for next step of measurement. I selected 2cm x 2cm fetal membrane close to the inner le-rvical oriface immediately after the labour of cases. All the samples were set in 10% formaldehyde, and embedded in paraffin.2, Using HE - staining, we could diagnose whether chorioamnionitis existed or not.3, Using immunohistological methods to test the quantity of NF - kB in all samples.4, Using ELISA (Enzyme -linked immunosorbent assay) to test the quantity of IL - 15 in pregnancy woman' s sera.5, Statistical analysis: Using SPSS software.
