ObjectiveThe cardiac sarcolemmal NCX1 and SERCA2a play an important role in the maintenance of the myocardial Ca2+ homeostasis which is altered in cardiac hypertrophy and failure. The present study used the STZ - induced diabetic rat to investigate the change of expression of the cardiac NCX1 mRNA and SERCA2a mRNA in diabetic cardiomyopathy and the relation in the change of NCX, mRNA and SERCA2a mRNA.MethodsExperimental diabetic rat was induced by streptozotocin (i. p. , 60mg/kg). After 4, 6 and 8 weeks, the myocardium of diabetic rats and that of control rats were isolated, respectively. Using RT - PCR technique we compared mRNA relative amount of NCX1 and SERCA2a in diabetic group and control group.ResultsThe weight of control group are significantly greater than those of diabetic group ( P < 0. 01 ) and the glucose levels increased significantly in diabetic group ( P <0.01). The ratio of the wet heart weight to the body weight of diabetic group is significantly greater than that of control group (P<0.05, P<0. 01). The relative amount of mRNA of NCX5 and SERCA2a in diabetic group is lower than that of control group in 4w, 6w and 8w group, but only the difference...
