Objective: To explore the expression of TRB3 in diabetic cardiomyopathy. To explore the relationship between TRB3 and apoptosis,interstitial fibrosis.Methods: 24 SD male rats were divided into control group(n=12)and diabetes group (n=12)randomly. Diabetes was induced by a single peritoneal injection of strepotozotocin (STZ,60mg/Kg). 8 weeks later, blood sugar and HbAc1 were tested.The changes of myocardium organization structure were observed by HE. Interstitial fibrosis was detected by Masson. Apoptosis of cardiomyocyte was determined by Tunel.TRB3 of each group was tested by qRT-PCR and Western Blot separately.Results: Compared with control group,there were such changes in diabetes group as the disorder and fracture of cardiocytes, and infiltration of inflammatory cells. There were more myocardial collagen content(P<0.01)and much more apoptosis of cardiomyocyte(P<0.01) in the hearts of diabetic rats. The expression of TRB3 in protein and mRNA was up-regulated(P<0.01). Significant positive correlations were observed between TRB3 protein and apoptosis(r=0.668,P<0.05), and between TRB3 protein and interstitial fibrosis (r=0.595,P<0.05).Conclusion: There are obviously increasing of protein and mRNA of TRB3 in diabetic cardiomyopathy.TRB3 may promote the development of diabetic cardiomyopathy by inducing apoptosis of cardiac myocyte andObjective: To observe the effect on the expression of TRB3 and apoptosis in HUVECs induced by high glucose.To explore its probable mechanism.Methods: After adjusting the concentration of glucose in culture solution,we divided the cells into different groups, as forrowing:group 5.5,group 15,group 25,group 35 and group 5.5,group 25,group 25+DMSO,group 25+SB203580.Apoptosis was detected by flow cytometry, TRB3 of each group was tested by qRT-PCR and Western Blot separately.Results: Compare with group 5.5,apoptosis and the expression of TRB3 were increasing .As the rise of glucose concentration, apoptosis and the expression of TRB3 were elevated gradually. The differences among groups were statistically significant.After pretreatment with SB203580 for 1h, apoptosis and the expression of TRB3 were blocked-up,but they were still higher than group 5.5.The differences between group 25 and group 25+DMSO on apoptosis and the expression of TRB3, were lack of statistically significant.Conclusion: High glucose can induce the expression of TRB3 and promote apoptosis partly through P38MAPK singal path at least.
