Objective:To investigate the Mechanism of the effect of Bushen Tiaojing Recipe on anovulatory disease,We observed the change of the level of vasorelaxation-vasoconstriction factors in plasma and studied the correlation of anovulatory disease and vasorelaxation-vasoconstriction factors.Discussed the effect of Bushen Tiaojing Recipe on vasorelaxation-vasoconstriction factors in ovary and its regulation and control function on sexual hormone,which offerred theoretical foundation to Bushen Tiaojing Recipe precipitating follicular development and ovulation in clinic application,it would make the development of gynecology of traditional Chinese medicine. Method:90 healthy female SD rats of 9-day age from the same batch whose weight were about 15g were adopted in our research.10 rats of them were made as normal group,which were injected 0.05ml neutral tea oil subcutaneously at nucha.Followed the methods of Wei Mei-juan,the other 80 rats were made as models by subcutaneous injection of testoserone at nucha.All these 90 rats were weaned when they at 22-day age. They were raised by Special messenger under the same condition and took food by freedom.When they were at 70-day old,their vagina debouched.We began to make the smear of vaginal epithelial cell for 2 successive sexual cycle(4-5 days per sexual cycle),Vaginal epithelial cell showed continuously cutinization,which hinted that models of anovulatory had set up successfully.40 female ASR were randomly divided into four groups,namely,group of Bushen Tiaojing high dose,group of Bushen Tiaojing low dose,group of Wuji Baifeng and model group,each group was made up of 10 rats.When the rats were at their 80-day old,the treatment groups were drenched with Bushen Tiaojing Recipe,the control group were treated with Wuji Baifeng oral liquid,the model and normal groups were treated with distilled water once a day continued for 7 weeks.While making vagina cells smear for 2 sexual cycles,recorded estrous cycle of each groups at detail.Fasting for 8 hours after administration for the last time.Decapitated the animals at preoestrus and collected their blood to prepare for plasma and serum.Measured the level of endothelin-1 (ET-1),angiotensin Ⅱ(AT Ⅱ),nitrogen monoxidum(NO),cyclic guanosine monophosphate(cGMP)as well as Estradiol(E2),Progesterone(P) and Testosterone(T)by radio immunoassay. We cut the belly open to take out the ovary on the right side under sterile operation,dehematized carefully and weighed,fixed them in 4% paraformaldehyde solution,paraffin imbeddinged on routine.We dyed them by HE and immunohistochemistryrespectively after serial sections in 5μm and observed morphological variation of ovaries and the expression intensity of ET-1, ATⅡ, nNOS,iNOS and eNOS. Results 1.Analysis of serum sexual hormone Compared with normal group,the level of serum E2,T and E2/P of model group significantly increased(P<0.01),after administrated with Bushen Tiaojing recipe,the level of serum E2,T and E2/P of anovulatory rats significantly decreased(P<0.01);There was no significant difference between the high-dose and low-dose groups(P>0.05);Compared with model group,the control group has the tendency to step down in the level of E2,T and E2/P,it has no statistical meaning(P>0.05);There was no obviously difference on P(P>0.05). 2.Analysis of vasorelaxation-vasoconstriction factors in plasma Compared with normal group,the level of ET-1 and ATⅡof model group significantly increased(P<0.01),while the content of NO and cGMP obviously decreased(P<0.01);after administrated with Bushen Tiaojing recipe,the level of ET-1 and ATⅡof anovulatory rats significantly decreased(P<0.01),and the content of No and cGMP increased(P<0.01),there was no obviously difference between the high-dose and low-dose groups)(P>0.05);There was no statistical difference between the control and model groups(P>0.05). 3.Analysis of vasorelaxation-vasoconstriction factors in ovaryCompared with normal group,the mean integral light density of the expression of granular cell of ET-1,ATⅡand iNOS in model group significantly inc...
