The Preparation Of Leishmania Antigen And The Research Of Application Value

Objective:To make and study the value of Leishmania promastigotes antigen from different strains in the immunity diagnosis byindirect enzyme-linked immunosorbent assay (ELISA).To exam Kala-azarpatients'serum and healthy persons' serum from epidemic area.To select out suitable strains to prepair for antigen, make serologic investigation result moreaccurate and reliable at Kala-field. Methods: 1.We collected Leishmania promastigotes,washed its with 0.01mol/LpH7.2 buffers ,smashed its,and then measured protein content and titrated work concentration(by ELISA).2.We tested Kala-azar patients'serum and healthy persons'serum of epidemic area by ELISA. The results of three antigens'methods were the same, work concentration of serum was the same(l :100).Results:The protein content of three antigens is, 901 strain with 1000ug/ml,801 strain with 960ug/ml and 951 strain with 1100ug/ml. Suitable work concentration is, 901 strain with 10ug/ml ,801 strain with 12ug/ml and 951 strain with 11ug/ml. 131 Kala-azarpatients were examined by ELISA with three Leishmania strains antigen, the results were 901 strain with 80.92%, 801 strain with 92.25% and 951 strain with 89.92%. The result was notable difference between 901 strain and 801 strain. 334 healthy persons serum of endemic areas were tested by ELISA with three Leishmania strains antigen, the positive rates were notable differenceamong three antigens. The positive rate of 901 antigen (10.48%) was higher than 801 antigen(5.99%) and 951 antigen(5.09%). There were notable different positive rates among three strains when we checked persons ofBACHU county, 901 strain was higher than 951 strain, but the results ofWUSHI county were not notable difference. The positive rates between men and women were not notable difference by ELISA with three strains antigen. The positive rates of three antigens on every age team were not notable difference. Conclusions: Leishmania promastigotes antigens have good stability and heat-resistant nature. The sensitivity of 901 and 801 antigen were better, so we thought it could be used for serologic epidemiology investigation in XinJiang. The specificity of 801 antigen was better, it could be used to exam specific antibody of Kala-azar and to study on Kala-azar.
