Objective: To study the mechanism of BuShenHuoLuoFang based on the role of invigorating the kidney treating female the sterility due to asthenia of the kidney and interpret the intervention role of promoting the growth of follicle and ovulation.Methods: Kidney - Deficiency Sterile Rats model was established by injecting testosterone propionate subcutaneously to SD female rats of 9 days age.Morphological and hormonal change of ovary and womb of Kidney-Deficiency Sterile Rats and Insulin-Like Growth Factor- I mRNA (IGF- I mRNA) expressions at its ovary in rats after feeding tonifying kidney herds(TKH) extract were observed by light microscope, in situ hybrizidation and radioimmuno-assay.Results: BuShenHuoLuoFang could increase the weight of ovary and womb of rat significantly and benefited the growth of follicle and ovulation. BuShenHuoLuoFang greatly increased the total of Ovum bubble quantity of different levels in ovary as well as the membrane thickness of womb andraised the levels of follicle-stimulating hormone (FSH) and luteinizing hormone (LH)in rat serum to improve the endocrine function and promote the growth of follicle and ovulation. BuShenHuoLuoFang significantly could strengthen the expression of the Insulin-Like Growth Factor- I mRNA (IGF-I mRNA) in rat ovary and favor the growth of follicle and ovulation.Conclusion: The results show that Kidney - Deficiency Sterile Rats model could be established by injecting testosterone propionate subcutaneously to SD female rats of 9 days age. Growth disorder and excretion disorder of follicle are important factor of the female sterility due to asthenia of kidney; BuShenHuoLuoFang based on the role of invigorating the kidney by affecting the level of FSH and LH of the rat improves its endocrine function and by affecting the expression of the IGF- I mRNA in ovary improves the growth of follicle thus achieving the purpose of promoting ovulation, which indicates that the treatment of Traditional Chinese medicine tonifying kidney herbs on kidney - Deficiency symptom truly has theory basis of molecale standard.
