Objective: To investigate the relationship between glucocorticoid receptor gene 2/1 codon 23 polymorphism and glucocorticoid-sensitive asthma. Methods: DNA was isolated from peripheral blood leukocytes using standard techniques. Polymerase chain reaction/Single-strand conformation polymorphism(PCR/SSCP) and gene sequence analysis technique were used in our study to detect and categorize the gene types of glucocorticoid receptor gene in exon 2/1 codon 23 of 20 asthmatic patients(asthmatic group) and 20 healthy people(control group). We analyzed the gene type frequencies of the two groups and estimated whether there was any difference between the two groups.We used X2-test to do statistical analysis.Results: PCR product of asthmatic group and control group length was 356bp. No aberrance was found in the glucocorticoid receptor genes of exon 2/1 codon 23 between the asthma group and the control group by SSCP. All gene types of the glucocorticoid receptor genes in exon 2/1 codons 23 were AGG.Conclusion: No correlation was found between glucocorticoid receptor gene in exon 2/1 codons 23 and asthma. Glucocorticoid receptor gene in exon 2/1 codons 23 may not be a key gene (susceptibility gene) in pathogenesis of asthma.
