The allograft heart transplantation has been developed to become an effective therapy for terminal heart disease. At present, donor heart preservation, surgical technique and the infection after transplantation have not been the major factors of the failure of transplantation. But the acute rejection still has serious influence upon the success of surgery and the survival of accepter. So, it is highly important to study the mechanism and the diagnostic method of the acute rejection. Many researches have indicated that there were many phenomena of microcirculation obstruction occurred during acute rejection of heart transplantation, such as, erythrocyte and leucocyte assembled around the cardiac muscle and vessel endothelial cell, many erythrocytes assembled in the blood vessel, leucocyte and platelet adhered to the vasal endothelium and lacuna vasorum was narrowed and blocked, etc. These phenomena implyed that the turbulence of cell hemorheology maybe play an important role in the course of acute rejection after heart transplantation. It is not clear of the action mechanism and the relation between the cell hemorheology and acute rejection. We supposed to observe the changes of cell hemorheology during acute rejection after heart transplantation to find out the relation.Objective: To observe the changes of cell hemorheology during acute rejection after heart transplantation and investigate the relationship.between acute rejection and cell hemorheology. To inaugurate a new field of acute rejection research.Methods:1. grouping: 6 healthy male SD rats were choiced randomly as normal group. Surgery trauma group(group A, 24 SD rats). Isograft group(group B, 48 SD rats) consisted of SD donors into SD recipients. Allograft group (group C, 24 SD rats and 24 Wistar rats) consisted of Wistar donors into SD recipients. The grafts were harvested at post operation days 1,3,5,and 7. The cell hemorheology of erythrocyte and leucocyte was measured , while pathologic examinations of the hearts were performed.2. Animal mode: Group A: The celiac artery and inferior vena cava of rats were interrupted after heparinization , and an incision was made into the vas then was sutured. After 30 minutes, the interdiction was opened. Group B and C: The model of rat heterotopic abdominal heart transplantation was used. The aorta and pulmonary artery of donor were connected with the celiac artery and inferior vena cava respectively by end-to-side anastomosis.3. Endocardium muscle biopsy (EMB): The grafts were harvested at post operation days 1,3,5, and 7d, and pathologic examinations of the hearts were performed.4. The erythrocyte deformability index (EDI), erythrocyte aggregation index (EAI), erythrocyte electrophoresis time (EET), and leucocyte deformability index (LDI) were measured at post operation days 1,3,5, and 7d. 5. The software program SPSS10.0 was used to deal with the experiment data.-5-Results :1. EMB : The acute rejection of group B was slight, and it of group C was aggravated gradually from 1st to 5th day after operation, and alleviated at 7th day post operation. The acute rejection of group C was significantly more severe than that of group B (P<0.001).2. The changes of the cell hemorheology : (D EET: There were no statistically difference between group A,B,C and normal group. (2) EAI: The EAI of group A and B was significantly increased at 1st day after operation, comparing with normal group, P<0.05, but there were no significantly difference at 3rd , 5th, and 7th day . The EAI of group C was significantly increased at 1st , 3rd, 5th and 7th day, and there were statistically difference with normal group, P<0.01. Group C was significantly different from group A and B, P.001. There was no statistically difference between group A and B. (3) EDI: There were no statistically difference between group A,B,and normal group.
