In this article, we studied the effects of Gossypium Herbaceum extract, Extract A and Extract B, prepared in two extraction methods, on coagulation time in mice and platelet aggregation and platelet adhererance and the formation of thrombosis in arteriovenous shunt model in rats, respectively. Extract A and Extract B were given at three doses (25 mg/kg,50 mg/kg,100 mg/kg) and Aspirin (24.7 mg/kg, ig) and Salvia Miltiorrhiza (105.3mg/kg,ig) were used as positive controls to evaluate whole coagulation time by glass slide method and the results showed that Extract A at a dose of 25mg/kg, i.g. markedly decrease the coagulation time (P < 0.05) while other doses having no significant effects on coagulation time. Platelet adherance function was assessed by using collogen adherance method with platelet adherance rate as the index. Extract A and Extract B were given at three doses (25 mg/kg, 50 mg/kg, 100mg/kg) with Aspirin (17.1mg/kg, ig) and Salvia Miltiorrhiza (72.9 mg/kg,ig) as the positive controls. The results showed that both Extract A at a dose of 25mg/kg and 50 mg/kg, ig and Extract B at at a dose of 25mg/kg, ig augmented the platelet adherance function in rats(P<0.05); Turbidity method was used to assess the platelet aggregation function inrats. ADP(Adenosine-5'-Diphosphate, final concentration was 3.33umol/l) and Col (Collagen, final concentration was 212ug/l) were used to induce platelet aggregation with Aspirin (17.1mg/kg, ig) and Salvia Miltiorrhiza (72.9 mg/kg,ig) as the positive controls, inducing platelet aggregation. Maximum aggregation rate was observed as the index. The results indicated Extract A (50 mg/kg,100mg/kg, ig) and Extract B(50mg/kg, 100 mg/kg, ig) had a marked inhibition effect on the ADP-induced platelet aggregation in rats (P<0.05 and P<0.001, respectively) . The inhibition rates were 16.79%,21.98% and 11.71%,17.67%, demonstrating dose-dependance. Extract A ( 100 mg/kg, ig) also showed a inhition for Col-induced platelet aggregation .The inhibition rates was 13.35%. Extract B (25mg/kg, ig) markedly shortened the latent phase of platelet aggregation induced by Col. Its latent phase was 1 minute compared with Aspirin, 3 minutes and Salvia Miltiorrhiza, 5 minutes.In the arteriovenous shunt model in rats, both Extract A (50 mg/kg, ig) and Extract B (50 mg/kg, ig) significantly decreased the wet weight of thrombus (P< 0.05) .The inhibition for thrombogenesis were 42.29% and 29.17%, respectively. Conclusion: The anti-thrombus effect of Extract A is more potent than Extract B and further optimization is needed on the basis of confirming the extaction method Extract A.
