The Experimental Study Of Jiaweishixiaosan On The Hemostatic Mechanism

Objective: In this research, the hemostatic effect and mechanism of Jiaweishixiaosan (JWSXS) were evaluated by observed the affect of JWSXS on the coagulation time , bleeding time and blood coagulation system, fibrinolysis system and platelet aggregation, the content of platelet a - membrance - coating granules protein, in under to provide the theoretic foundations for the clinical applications and the exploitation of new herbal medicine preparation.Methods: After ig JWSXS 5.0-10.0g.kg-1.d-1, P-Aminomethyl Benzoic Acid(PAMBA, as the positive control ) 0.05g.kg-1.d-1 for six deys in mice, thecoagulation time (CT) was measured by capillary method and the bleedingtime (BT) was measured by cutting tail method. After ig JWSXS 4.0-8.0g.kg-1.d-1, PAMBA 0.04g.kg-1.d-1 for six deys in rats, the prothrombin time(PT) and the kaolin partical thromboplastin time (APTT) were measuredby the ACL-200 antomatic bood agglutination instrument . The activities ofthe tissue plasminogen activator (t-PA) and the plasminogen activatorinhibitor-1 (PAI-1) were measured by ELISA method. After ig JWSXS3.0-6.0 g. kg-1.d-1 for six deys in rabbits, the BS-631 blood-plateletagglutometer was applied to determination of platelet aggregating function, while the enzymelinked immunosorbent assay (ELISA) method adopted to observe the content of a -membrance-coating granules protein (GMP-140). Result: JWSXS in twain dosage and PAMBA could obviously reduce the blooding time, the coagulation time, the prothrombin time, the activated partical thromboplastin time and the activity of the tissue plasminogen activator and increase af the activity of the plasminogen activator inhibitor-1. The results ware compared with normal contral group, and have the significent difference (P<0.01). In the meantime, there were no any effect on the platelet aggregating function and the content of a - membrance - coating granules protein, compared with normal contral group, and have not the significent difference (P>0.01).Conclusions: (1) JWSXS in twain dosage and PAMBA could all come into being the hemostatic effect. Both JWSXS and PAMBA is resemble in the character of effect but the effect of big dosage JWSXS is much strong. (2) The result is suggested that the hemostatic mechamism of JWSXS is to stimulate the intrinsic and extrinsic blood coagulation system and inhibit fibrinolysis system. JWSXS is mostly effect to the blood coagulation system, PAMBA is mostly effect to the fibrinolysis system. (3) Neither twain dosage JWSXS is effecting the function of the platelet aggregation, nor effecting the plateleta-membrance - coating granules protein,which is probably the characteristic of the drug arresting of Weeding by removing stasis.
