Objective: To observe the change of cytokines gene expression in pathological rat before and after celiac infection.Methods: Three animal models of control, liver cirrhosis and immunosuppression were established in 90 Sp -ague-Dawley rats, each rat was carried out caecal ligation and puncture (CLP), TNF-a and IL-6 gene expression in liver and lung tissue were measured with reverse- transcription polymerase chain reaction (RT-PCR).Results: 1. Before CLP, TNF- a mRNA and IL-6mRNA in liver cirrhosis group were higher than that of the control group(P<0.01). Otherwise, cytokines gene expression in immunosuppression group was lower than that in the control group(P<0.05). 2. After CLP, c>tokines gene expression in each group changed similarly. That was, TNF- a mRNA and IL-6mRNA in different tissue began increasing at the 3rd hour (P<0.05), then increased smoothly.3. After CLP, TNF- a mPNA and IL-6mRNA in liver cirrhosis group were always higher than that of the control group(P<0.05),but cytokines gene expression in immunosuppresiiion group was lower, especially in TNF- a mRNA of the lung tissue(P<0.01).4. In liver cirrhosis group, theaverage increased multiple of IL-6mRNA after CLP was 7.45, but that of TNF- a mRNA was 3.86. In immunosuppresj ion group, that were 8.97 and 3.96.Conclusions: 1. In pathogenic condition, the change of TNF- a and IL-6 in rat before and after celiac infection probably happens at the cytokines gene transcription level. 2.Before CLP, immune function of liver cirrhosis rat has already been disordered, this may relate to the forming of liver cirrhosis .3. After CLP, TNF- a mRNA and IL-6mRNA in liver cirrhosis rat go on increasing at the higher level, this may cause multiple organ dysfunction syndrome (MODS) more easily. 4. Immuiosuppressive rat with celiac infection may develop into MODS, the low expression of cytokines probably due to compensatory anti-inflammatory response syndrome (CARS). The change of cytokines gene expression in this condition is similar to the change in MODS caused by systemic inflammatory response syndrome (SIRS). 5. In the course of infection, IL-6mRNA change:; more sensibly and can be a prognostic indicator in severe infection.
