Objectives: Our aims were to investigate effect of taurine on interfering in myocardial fibrosis and to discuss the potential mechanism via isoproterenol induced myocardial fibrosis rat model.Methods: 36 male Sprague-Dawley rats were divided into 3 groups randomly and equally: Iso group, taurine-treated group, and control group. Rats in Iso group were daily subcutaneously injected with isoprenaline (0.5 mg.kg-l.d-1) for 10 days; Rats in taurine-treated group were treated with taurine 300 mg.kg-l.d-1 after injections with isoprenaline; Rats in control group were daily subcutaneous injected with isoprenaline saline at the same dose. Every group was divided into 2 subgroups in 10th and 30th day respectively. All rats were anaesthesiaed and their hearts were quickly taken out and washed with cold NS, then dried by filter paper and weighed. The ratio of heart weight to body weight, left ventricular mass index were figured out. In left ventricular myocardial, we measured the expressions of collagen type I, III and CTGF mRNA with RT-PCR, detected CVF with Van Gieson Collagen staining and used immunohistochemistry to determine CTGF levels.Results: 1-. The ratio of heart weight to body weight and left ventricular mass index were significantly increased on 10th and 30th day after subcutaneous injections with isoprenaline. There were significant differences among the three groups. 2, Compared with control group after the injections, cardiac collagen types I, III mRNA and CTGF mRNA were significantly increased at lOd and 30d in Iso group and taurine-treated group. There were significant differences between Iso group and taurine-treated group as well.Conclusions: 1 -. Low dose of isoprenaline could lead to myocardial fibrosis, with leftventricular changes obviously. And this myocardial flbrosis was observed at 10th and 30th day. 2, CTGF is involved in the course of myocardial flbrosis, and correlate to collagen type I, III mRNA. 3, Taurine can hold back CTGF and collagen type I, III mRNA expression , decrease ratio of heart weight to body weight, LVMI. Therefore it can prevent myocardial flbrosis.
