Berberine With The Taurine Ir Rats With Early Intervention And Adipose Tissue Of Pai-1 Mrna Expression Experimental Study

Objective To observe the effects of berberine and taurine on PAI-1 mRNA expression in epiploon adipose tissue of high fat rats model of insulin resistance(IR). and to study their effects on blood glucose, blood lipids, insulin levels, insulin sensitivity index(ISI)and body weight. Methods SD rats were divided into normal and model groups at random by weight;model groups were fed with high fat diet for 30days firstly, screened the rats whose ISI decreased as model rats. 40 IR rat models induced by high fat diet of 30 days were randomly distributed into five groups(8 rats/group): berberine group;taurine group; berberine combined with taurine (combine)group; metformin group;model group. After treatment,using RT-PCR to PAI-1mRNA from adipose tissue,the products were obtained,semi-quantitative RT-PCR employed to study its expressions in adipose tissues of rats with different groups. The fasting plasma glucose(FPG),insulin(FINS),total cholesterol(TC),triglycerides(TG), high–density lipoprotein cholesterol(HDL-C),Low-density lipoprotein cholestero(LDL-C), free fatty acids(FFA),and lipocyte area of internal organs were performed in each groups.ISI was calculated. Results (1)The IR model rats were induced by high fat diet (59% calories from fat). FPG,FINS,TG,TC,LDL-C of Model group were increased,ISI were decreased . The expression of PAI-1mRNA of adipose tissues was increased significantly (.2)Compairing with model group,the berberine and taurine group and combine group could significantly increase insulin sensitivity, decrease the expression of PAI-1mRNA in epiploon adipose tissue. The parameters mentioned above in combine group was better than other groups. Meanwhile the secretion of FFA and lipocyte area of internal organs decrease.(3)The decrease of TG,TC ,LDL-C and the increase of HDL-C in combine group and taurine group were better obviously than other groups. Conclusion Application of combining Berberine and taurine is effective in increasing insulin sensitivity IR in rats induced by high-fat diet and decreasing the expression of PAI-1-1mRNA in adipose tissues.It had significant effect on changing the total status of insulin resistance.
