Object To explore the changes of diabetic retinopathy-related factors MMP2,Collagen I,and CTGF after the action of delphinidin on HFSF cells.Methods Resuscitated and cultured HFSF cells with delphinidin at 0,10-6,10-5,10-4,10-3,10-2,10-1,1 mg/mL.MTT assay was used to detect the proliferation of HFSF cells,and the cell morphology and density of each group were recorded under inverted optical microscope after 8h,16h,24h and 48h.Setting up control group?0 mg/mL?and delphinidin group?10-11 mg/mL?for48h,test cell cycle and apoptosis?by flow cytometry?,MMP2?COL1A1?CTGF genes?by qRT-PCR?,MMP2?Collagen I?CTGF proteins?by Western Blot?.Results The HFSF cells cultured in vitro were triangular and star-shaped?arranged in a network for 8h,the cell density increased?mostly be fusiform and bundled arranged after 48hours under inverted optical microscope.The cell density decreased and the morphology changed after treated with delphinidin.MTT showed that the proliferation index of HFSF cells changed with different concentrations of delphinidin for different times?P?0.05?.The HFSF cells were more severely inhibited with higher concentration of delphinidin.Flow cytometry showed:the percentage of cells in G1 phase of the blank control group and 10-1mg/mL group were?65.23±0.59?%??69.92±0.66?%,in the?S+G2?phase were?34.76±0.59?%??30.08±0.66?%?P?0.05?;The early apoptosis rate were respectively?4.02±0.82?%??22.97±0.2?%?P?0.05?,late apoptotic rate were?3.58±0.10?%??3.57±1.47?%?P?0.05?.The gene expression of MMP2,COL1A1 and CTGF decreased tested by qRT-PCR?P?0.05?;The protein expression of MMP2 and CTGF decreased?P<0.05?,but the expression of Collagen I protein was not statistically significant by Western Blot?P?0.05?.Conclusion Delphinidin inhibits the proliferation of HFSF cells in vitro and reduces the expression of DR related factors MMP2?CTGF in HFSF cells.
