Objective: To investigate the changes of serum IL-1β,TNF-a and IL-8 inpatients with acute pulmonary thromboembolism (PTE) and the relationshipbetween these cytokines and the disease.Methods: Radioimmunoassay was applied to measure serum levels of IL-1β,TNF-a and IL-8 in 18 acute pulmonary thromboembolism patients (11 massive, 7minor ) in different time. 20 patients undergoing the fracture of femur bone (oneof risk factors for PTE) and 18 healthy subjects were selected as two controlgroups.Results: (1) on the day of admission, the median concentrations in serum of IL-1β,TNF-a and IL-8 wereï¼ï¼Žï¼—228±ï¼ï¼Žï¼’654 ngï¼ï½ï½Œ,1.4ï¼ï¼™ï¼”±ï¼ï¼Žï¼”ï¼’ï¼ï¼— ngï¼ï½ï½Œ,1.345±ï¼ï¼Žï¼“795 ngï¼ï½ï½Œ,respectively, in the PTE group ; 0.4025±0.1912 ng/ml,1.1415±0.3268 ng/ml,0.9056±0.3308 ng/ml, P<0.05, respectively, in the fracture group ï½ï½Žï½„ ï¼ï¼Žï¼’228±ï¼ï¼Žï¼‘594 ngï¼ï½ï½Œ,ï¼ï¼Žï¼™ï¼ï¼•ï¼–±ï¼ï¼Žï¼’923 ngï¼ï½ï½Œ,ï¼ï¼Žï¼—728±ï¼ï¼Žï¼“365 ngï¼ï½ï½Œ, P<ï¼ï¼Žï¼ï¼ï¼ï¼‘,respectively, in the healthy group. (2) During the next days, IL-1βconcentration showed a gradual decline in patients with PTE from ï¼ï¼Žï¼—228±ï¼ï¼Žï¼’654 ngï¼ï½ï½Œã€€ï½”ï½ã€€ï¼ï¼Žï¼’688±ï¼ï¼Žï¼‘656 ngï¼ï½ï½Œ,  (P <.001), TNF-a concentration declinedfrom 1.4ï¼ï¼™ï¼” ± ï¼ï¼Žï¼”ï¼’ï¼ï¼— ngï¼ï½ï½Œ to 1.ï¼ï¼‘13 ± ï¼ï¼Žï¼’892 ngï¼ï½ï½Œ,(P<ï¼ï¼Žï¼ï¼ï¼‘),ï½ï½Žï½„interleukin-8 concentration was relatively constant in time,  frï½ï½ã€€ï¼‘.345±ï¼ï¼Žï¼“795 ngï¼ï½ï½Œã€€ï½”ï½ã€€ï¼‘.1575±ï¼ï¼Žï¼“ï¼ï¼—9 ngï¼ï½ï½Œ,(P>ï¼ï¼Žï¼ï¼•).(3) Seruï½ã€€ï½Œï½…vels ï½ï½†ã€€IL-1β,TNF-a and IL-8 in ï½ï½ï½”ients with ï½ï½ƒï½•ï½”e ï½ï½ï½“sive PTE were higher thï½ï½Žã€€ï½”hï½ï½“e with ï½ï½‰ï½Žï½ï½’ (0.8609±0.2346 ng/ml vs 0.5057±0.1334 ng/ml, 1.5855±0.4145 ng/ml vs 1.1329±0.2625 ng/ml, and 1.5527±0.3484 ng/ml vs 1.0186±0.4741 ng/ml, respectively, P<0.001).Conclusion: (1) IL-1β,TNF-a and IL-8 may play roles in the pathogenesis ofAcute pulmonary thromboembolism. (2) The association between elevatedIL-1β,TNF-a and IL-8 concentrations and the acute phase of PTE provides IIIevidence for a role of inflammation in thrombosis. (3)Such level of elevation ofIL-1β, TNF-a and IL-8 may be involved in the development of venousthrombosis among patients with leg fracture.
