| Objective: To study NGF, BDNF and NT3 expression changes in different interval after transected spinal cord in rats.Methods: Total 25 rats were randomly divided into 5 groups: Normal group, after trans-section spinal cord operation for 3days, 7days, 14days and 21 days groups. We observe the expression changes of NGF, BDNF, NT-3 in different interval after transection operation in rats by ABC immunohistochemical staining and HPISA-1000 High Resolution Pathological Image Analysis System.Result: 1). NGF positive cells could be observed in dorsal horn and ventral horn in normal spinal cord. And NGF immunoreaction product was located in plasma and nucleus and most in plasma. NGF positive cells were increase after trans-section operation compared with normal group, notably in 7 days group (P<0.05). 2). BDNF positive staining was confined to locate in plasma of motor neurons in ventral horn. BDNF positive cells in 3 and 7 days groups after operation were also increased compared with normal group, significantly in 7 days group (P<0.05). While there was not difference between 14 days, 21 days groups and normal group in dorsal and ventral horn neurons. 3). NT3 immunoreactivity was in plasma and nucleus of dorsal and ventral horn cells. The NT-3 positive cells number was markedly increased after operation compared with normal (PO.05). Between every two groups of different interval days after operation treatment, the NT-3 positive cells number was noticeable difference too (P<0.05). 4). The mean gray levels of NGF, BDNF and NT-3 were reduced in different interval days after operation compared with normal in ventral and2dorsal horn area.Conclusion: NGF, BDNF and NT-3 positive neurons was increased in ventral and dorsal horn after transection operation treatment in rats and mean gray level was decreased compared with normal group, which has shown that expression changes of NGF, BDNF and NT-3 maybe is relation to spinal cord plasticity. That NGF and NT-3 positive cells numbers in different interval days after transection operation was increased suggest expression of NGF and NT-3 was sustained enhance. While expression of BDNF was only transitorily increase. |
