| Objective To study the effect of composite Triptcrgium Hypoglaucum hutch(CTHH) on the intercellular adhesion molecule( ICAM-1) expression on human vascular endothelial cells . Methods The effect of the CTHH on TNF-alpha induced ICAM-1 expression on human vein endothelial cells (HUVEC, ECV-304 cells) was measured by immunohistochemistry . Results 1. Compared with blank control, CTHH at dose of 0.05-2mg/ml down-regulated TNF-alpha induced ICAM-1 expression (P<0.05),CTHH at dose of 2mg/ml inhibited ICAM-1 expression more significantly (P<0.01). 2. Compared with TriptergiumHypoglaucum(TH), there is no difference on inhibiting the ICAM-1 expression between CTHH and TH (P>0.05). 3. Compared with dexamethasone(DM. lug/ml), there is no difference on inhibiting the ICAM-1 expression between CTHH at higher dose (0.5-2mg/ml) and DM ( P>0.05) ,but the ICAM-1 expression on the HUVEC that were treated with DM is less than the CTHH at the lower dose (0.05mg/ml) (P<0,05). Conclusion CTHH (0.05-2mg/ml) decrease the ICAM-1 expression on vascular endothelial cells in a dose-dependant manner . It indicated that down-regulated expression of ICAM-1 and adhesion between leukocyte and endothelial may be the possible therapeutic anti-inflammatory mechanism of CTHH . |
