Objective To study acute toxicity ,pharmacod/namics and mechanisms of the composite triterygium hypoglaucum hutch (CTHH) to suppress effectiveness on photoallergic reaction of murine and to compare with the single THHs (STHHs) that extract from water and 95% ethanol and non-THH composite. Method To observe ICR murine toxic by oral ing the CTHH, the STHHs that extract from water and 95% ethanol and non-THH composite; The photoallergic animal model of Balb/c murine was established upon using photosensitizer chlorpromazine and UVA irradiation. The efficacy of each experimental group was determined with measuring the murine's ear swelling thickness,weight difference and quantity of IFNiltration monocytes by pouring medicine to murine's stomach. The medical mechanism of composite THH were expounded by measuring ICAM-1 expression in keratinocyte, dermal interstitial cell and perivascular cell by immunohistochemical technology and by measureing IFN-γ quantity in serum by ELISA technology. Results The LD50 of the STHHs that extract from water and 95% ethanol are 78. 02g/kg and 35. 57g/kg. The LD50 of the CTHH is not able to acquired and it' s maximum tolerance dose (MTD) is 439. 20g/kg , be equal to the STHH 86. 97g/kg. The MTD of non-THH composite is 380. 95g/kg. The differences of murine's ear swelling thickness, weight and quantity of IFNiltration monocytes that was treated with the CTHH were significant less than the controls of the STHH that extract from water and alcohol and non-THH composite (P<0.05) as well as the controls of 0. 9%NS(P<0.01). The ICAM-1 expression on skin and IFN-Y quantityin serum that were treated with composite the THH were also obvious lower than the control of 0. 9%NS (P<0.05) . Conclution The CTHH acute toxicity is less than the STHHs .The CTHH has markedly suppressive effects to photoallergic reaction and is more effective than STHHs and non~THH composite. The partial mechanism is suppressed ICAM-1 expression in skin and lymphocytes producting IFN- γ so as to reduce IFNiltration of monocyte and to relieve allergic reaction.
