The invasion and metastasis of human colorectal carcinoma is a complicated process in which the signal of the carcinoma cells across membrane transferring and the interaction of the carcinoma cells and ECM play an important role to the proliferation, adhesion, invasion and metastasis of the carcinoma cells. Integrin alpha5betal is an important membrane receptor of the cell, its binding to his specific ligand FN play an important role to the proliferation, adhesion, differentiation invasion and metastasis of the carcinoma cells, the ion exchange and signal transferring. The changes of the expression of Integrin alpha5betal, FN and the component of signal transferring gateway can influence the biologic behavior of the colorectal carcinoma.To study the correlation of FN and integrinasB1 with colorectal carcinomas, the expression of the integrin alpha5betal and FN was detected by Fluorescence quantitive polymerase chain reaction and immunohistochemistry in normal colonic epithelium and carcinoma from the protein and transcription level. The results indicated that integrin alpha5betal is significantly higher expressed in colorectal carcinoma compared with normal colonic epithelium (P<0.05). the expression of the integrin alpha5betal had relation with histological grade and invasion degree (P<0.05). the expression of the integrin alpha5betal in moderate or well differentiation colorectal carcinomaswas lower than that of the poor. The expression in invasion profound carcinoma is higher than in invasion shallow carcinoma, the expression of integrin alpha5betal in coiorectal carcinomas wasn't related with the lymph node metastasis (p>0.05). these data demonstrate the expression integrin alpha5betal had closely correlation with carcinogenesis, higher invasion competence and poor differentiation tendency of coiorectal carcinoma, the data of immunohistochemistry detected suggested that the expression of FN in coiorectal carcinoma be significantly higher than that of normal colonic epithelium (P<0.05), but not relation with the histological grade, the invasion degree and the lymph node metastasis (p>0.05). the data of FQ-PCR detected indicated that the expression of FN in coiorectal carcinoma was significantly higher than that of in normal colonic epithelium and had relation with histological grade (P<0.05), but not relation with the invasion degree and the lymph node metastasis (p>0.05). with the method of collation analysis to prove integrin alpha5 subunit had positive correlation with FN in the gene transcription level (P<0.05) .Our results suggested that the high expression of the integrin alpha5betal and its ligand FN in protein level is the result of up-regulated in the gene transcription level simultaneously. The expression of the integrin alpha5betal had closely correlation with carcinomatous change, higher invasion competence and poor differentiation tendency of coiorectal carcinoma. In conclusion, the research indicated that the integrin alpha5betal and FN have a significant meaning in prognosis and treatment of the coiorectal carcinoma.
