The reject action is the key issue to affect success rate in the organ transplantation.With the immunosuppression drug's unceasing renew and reasonable usage,the success rate of organ transplantation and existing time after operation are gradually rising. But most patients are also hard to bear the cost of expensive drug and various kinds of ill effects,the most capital question is that immuno-suppression drug can't solve the chronic rejection of graft.In the last,the graft gradually miss the function.To solve the graft rejection thoroughly,the key point is to induce none of immune response to donator's organ in recipient,it is called immune tolerance. As a special immune organ,the role of liver in induction immune tolerance is gradually known by us.It is clear in some investigation,the selective catch of Kupffer cells in recipient liver to lymphocytic of donator in the way of injection of vein,is the key issue to induce and maintain immune tolerance through portal vein.The theory had been confirmed in the experiment with small animals,but none test have been done by using large mammalia.We made dogs as our object in the research and done as following: Objective:Using dogs as our investigative object,observe whether it can induce immune tolerance in dogs that received donor spleen cells through portal vein after renal transplantation.Method: 1.Animal dividing groups:Team A Contrast team:five dogs will simply be proceed kidney transplant, offer spleen cells and kidneys which will be transplanted to experiment B and C teams.Team B Portal vein injection team:five dogs will be injected donor's spleen cells through portal vein at the five to seven days before kidney transplant.Team C CsA team:five dogs will be given CsA 10-15mg/kg.d after kidney transplant.Team D five dogs provide kidney to contrast team for kidney transplant. 2. Preparing spleen cells: Spleens were got with aseptic manipulation,milled it in 100 pore steel wire,centrifugation after collecting cells, dissolved erythrocyte with Tris-NH4Cl after remove superior pure fluid,adjusted the number of cells to 2*107ml-1 with RPMI1640.3. Injection donor spleen cells through portal vein and renal transplantation: Abdominal cavity of B team dogs were opened, suspense fluid of spleens was drew out 0.5ml with injection syringe and injected along hepatic portal vein.Renal transplantation was proceeded after 5-7days.Low molecular disebrin was injected to dogs of A and D teams before transplantation and the kidneys out of body were filling with preservation solution.The kidneys were transplanted to dogs' iliac fossa of B and C teams, ureteral orifices were exposed in vitro.One kidney of D team dogs were transplanted to the iliac fossa of A team dogs in the same way.CsA was added to C team dogs' foods according to 10-15mg/kg each day after transplantation.Results:1.Result of renal function:The renal function of the dogs before operation had no significant difference;but renal function of the control team was much higher than B and C teams,and the difference was notable;difference of blood creatinine between B and C teams was not significant.2.The survival time of each teams after operation:The dogs of B and C teams had longer survival time than control team,and the difference was notable;but the survival time between B and C teams was close to each other,the difference was not significant. 3.Determinations of IL-2 and IL-6 in dog's serum after operation:The concentration of IL-2 and IL-6 in control team was much higher than B and C teams;but the difference between B and C was not significant.4.Pathology check result:In the light microscope,there are kidney acute rejection pathologic alteration such as kidney interstitial edema and lymphocyte infiltration in the control team,but in the B and C teams ,there are glomerular capillaries dissepiment shrinkage gently and basement membrane thicken.Discussion:As the maximum Reticuloendothelial cell systerm,liver can phagocytize foreign antigen.It has been certified...
