ObjectsTo study and compare the level of resistin in normal control subjects and type 2 diabetic patients, to assess the association of obesity and insulin resistance with resistin.MethodsELISA was used to examine the fasting plasma resistin in 43 type 2 diabetic patients,27 males and 16 females,and41 normal control subjects, 24 males and 17 females. Height, weight, waist circumstance and hip circumstance were measured. Body mass index (BMI) and waist to hip ratio ( WHR) were calculated. Fasting plasma glucose (FPG) ,triglyceride(TG) , total cholesteryl (TC) , high density lipoprotein ( HDL) and low density lipoprotein ( LDL) were examined in central laboratory. Fasting insulin ( FINS) was examined in endocrine laboratory. Insulin sensitivity index (ISI) was calculated.Result1. In the two group, the fasting plasma resistin concentrations were significantly higher in obese compared with lean subjects ( P < 0. 05 ). The fasting plasma resistin concentrations was positively correlated with BMI(r =0. 23 ,P < 0.05).2. In comparison with the lean control, the lean diabetes had higher fasting plasma resistin concentrations ( P < 0. 01). The fasting plasma resistin concentrations were significantly higher in obese diabetes than obese control (P < 0. 05) . Resistin was negatively correlated with ISI(r= -0.31,P<0.05).3. Resistin was negatively correlated with FINS (r= - 0. 34, P < 0.05 ) and not significantly correlated with FPG.4. Resistin was not significantly correlated with Serum lipid indexes, age , sex and WHR.5. The equation was calculated by regression analysis [ y = -0. 978 +0. 055x3 - 1. 387x6 - 1. 071x7i resistin ( y) , age (Xi) , sex (x2), BMI ( x3) , WHR ( x4 ) , FBG( x5) , FINS ( x6 ) , ISI ( x7) ]. So fasting plasma resistin was positively correlated with BMI and negatively correlated with FINS and ISI.DiscussionResistin is a novel adipocyte - derived polypeptide hormone and was found by Steppan et al in 2001. The resistin is a disulfide - linked homodimer and conformed by three domain-the N - terminal signal peptide containing 20 amino acid residues, the highly variable middle domain and the C - terminal sequence containing serine and cysteine residues with a unique cysteine repeat motif of CX (12)CCX(12)CX (8)CXCX(3) CX(10)CXCXCX(9)CC.Resistin levels are markedly elevated in mice that developed obesity and insulin resistance fed on a high - fat diet and also elevated in ob/ob and db/db mice in which obesity and diabetes are inherited traits. Blood glucose levels increase after intraperitoneal administration of recombinant resistin. Neutralization of resistin reduce the hyperglycaemia of obese, insulin - resistant mice, at least in part, by improving sensitivity to insulin. Resistin is thus a hormone that potentially links obesity to diabetes. We studied and compared the level of resistin in normal control subjects and type 2 diabetic patients by ELISA. In the two group, the fasting plasma resistin concentrations were significantly higher in obese compared with lean subjects (P <0. 05). The fasting plasma resistin concentrations was positively correlated with BMI (r = 0. 23,P<0. 05). These suggested that human fat tissue was able to excrete resistin. The more fat tissue body has, the more resistin fat tissue excretes. We also found resistin was not significantly correlated with WHR. These suggested resistin was correlated with the amount of fat tissue and not correlated with the distribution of body fat.Insulin sensitivity index is an index which reflect insulin resistance in vivo. The less insulin sensitivity index is, the more severity insulin resistance. We found resistin was negatively correlated with ISI(r= -0.31,P<0. 05). These suggested that the higher resistin levels, the more severity insulin resistance. Our results showed in comparison with the lean control, the lean diabetes had higher fasting plasma resistin concentrations( P <0.01). The fasting plasma resistin concentrations were significantly higher in obese diabetes than obese control ( P <0.0... |
