| Objective Obesity is a disorder that increases the risk of insulinresistance,Adipose tissue plays an active role in the regulation of fuel metabolismand energy homeostasis by expressing a series of regulatory genes, for example:peroxisome proliferators activated receptor gamma 2,leptin and tumor necrosisfactor-α.We study their relationships among these factors and their associationswith insulin resistance. Methods Fasting serum levels of leptin, insulin,glucosewere measured in 17 obese and 21 nonobese subjects. The mRNA expression ofPPARγ2, leptin, TNF-αof subcutaneous adipose tissue in the abdomen wasquantified in all of subjects by RT-PCR. Insulin resistance index(IRI) wascalculated in Homeostasis model assessment, HOMA). Then analyze therelationships between them. Results Y The mRNA expression of PPARγ2leptin and TNF-αin the subcutaneous adipose tissue were significantly higher inobese patients than in nonobese subjects(PPARγ2 mRNA: 1.532±0.80 vs 0.96±0.45,P<0.05; leptinmRNA: 1.17±0.73 vs 0.80±0.20 P<0.05; TNF-αmRNA:1.34±0.60 vs 0.75±0.34;<0.001) T P . The fasting serum levels of leptin ,insulinand IRI were significantly higher in obese patients than in nonobesesubjects(FINS:26.129±13.14 mU/L vs 8.227±3.34 mU/L,P<0.001;leptin:10.14±4.68 ng/ml vs 5.79±1.66 ng/ml,P<0.01;IRI: 5.43±6.11 vs 1.51±0.84,P<0.001). ? PPARγ2mRNA, leptin mRNA and TNF-αmRNA levels correlatedwith each other(PPARγ2mRNA and leptin mRNA:r=0.438,P<0.01; PPARγ2mRNA and TNF-αmRNA:r=0.541,P<0.01; leptin mRNA and TNF-αmRNA: r=0.578,P<0.001) and all correlated with IRI, BMI,serum levels of leptin andinsulin. à IRI correlated with PPARγ2 mRNA,leptin mRNA,TNF-αmRNAlevels, serum levels of leptin and BMI; Serum levels of leptin correlated withPPAR? mRNA leptin mRNA TNF-αmRNA levels, IRI and BMI,but not 2 4correlated with fasting glucose ; a Further analysis revealed that IRI,BMIcontributed to the degree of PPAR? 2 mRNA(PPARγ2 mRNA=-1079+0.043BMI+0.237 IRI,r2=0.459,P<0.001);BMI and leptin mRNA contributed to thedegree of TNF-αmRNA(TNF-αmRNA=-0.870+ 0.036 BMI+0.487 leptinmRNA,r2=0.573,P<0.001); TNF-? mRNA contributed to the degree of leptinmRNA(leptinmRNA= -0.022+0.612TNF-αmRNA,r2=0.433,P<0.001); PPARγ2 mRNA contributed to the degree of IRI(IRI=0.358+0.813PPARγ2mRNA,r2=0.317,P<0.001);,BMI contributed to the degree of serum levels of leptin(Y=-0.525+0.559BMI r2=0.270,P<0.01).Conclusion Y The mRNAexpressionof PPARγ2,leptin, TNF-? subcutaneousb adipose tissue in abdomen weresignificantly higher in the male obeses. T Leptin mRNA, TNF-αmRNA influenceeach other. ? BMI,IRI were important factor in influence of the mRNA expressionof PPARγ2 in males. PPARγ2 was one of valuable indexes in evaluating insulinresistance. Serum levels of leptin correlated with leptin mRNA levels.BMI was themost important factor in influence of serum levels of leptin. |
