| Objective To compare the effects of Dobutamine accompanied with Meglumine cyclicadenylate(MCA) or Milrinone on the plasma angiotensin(Ang II ),intraerythrocytic Ca2+ concentration and the myocardial contractility and relaxation of left ventricular in experimental rats with heart failure (HF).Methods The wistar rats model with HF was produced by injecting intraperitoneally adriblastina(ADR).The 40 rats were randomly divided into four groups(l)HF controls,(2)Dobutamine, (3)Concomitant Dobutamine and MCA,(4) Concomitant Dobutamine and milrinone groups,and (5) normal group was selected randomly to serve as control. After 10 days of medical therapy, hemodynamic studies Ang II and RBC-Ca2+ contents were performed in each group.Results In group Dobutamine +MCA, ±dp/dtmax, LVSP increased and LVEDP decreased (P<0.05-0.01). Ang II and RBC-Ca2+ contents decreased. In group Dobutamine+milrinone,+dp/dtmax and LVSP increased (P<0.05-0.01). Ang II decreased(P<0.05).In group Dobutamine+ MCA, LVSP,LVEDP, -dp/dtmax, Ang II and RBC-Ca2+ contents were significant differences in comparison with those in group Dobutamine (all P<0.05).Conclusion Both Dobutamine accompanied with MCA and Dobutamine accompanied with Milrinone can reduce the plasma concentration of Ang II and improve the myocardial contractility of left ventricular in HF rats. Dobutamine accompanied with MCA can also improve myocardial relaxation of left ventricular and reduce RBC-Ca2+ contents in HF rats. |
