| Thermodynamic Studies on the Chinese Materia MedicaFour Natures of Preparations and Compatibilities of Coptidis Rhizoma and its active essencesFour Natures of Traditional Chinese Medicine (TCM) is the foundation and core of TCM. Guided by the theories both TCM and Thermodynamics, Thermodynamics of TCM was first put forward and was introduced into the study of Four Natures. In this paper, mainly by the biothermoeffect evaluation method--Microcalorimetry, the evaluation method for Four Natures of TCM was established preliminarily. This method together with fingerprint chromatography and computer pattern-identification, Four Natures variation and active essences of Coptidis Rhizoma and its preparations, the compounds combined Coptidis Rhizoma with Evodiae Fructus were explored.Results and conclusions as follows:1. Microcalorimetry was first used to obtain the metabolic growth power-time curves of bacteria and biothermokinetic parameters in different conditions. Combined with the records of ancient herbal literatures, the natures of the preparations of Coptidis Rhizoma(Jiangzhihuanglian, Wuyuhuanglian, Jiuzhihuanglian, Cuzhihuanglian and Danzhihuanglian) and the compounds combined Coptidis Rhizoma with Evodiae Fructus (Fanzuojinwan, Zhuyuwan, Ganlusan and Zuojinwan) were synthetically evaluated. All the samples could restrain the bacteria growth and metabolism in different levels. Drugs with Hotter Nature could make the organism decreasing the multiplication rate constants and increasing heat output(AH) remarkably, while drugs with Colder Nature could make the organism decreasing the multiplication rate constants and increasing heat output(AH) a little. There was a stable difference between them. So the total heat output( A H) could be the objective parameters to evaluate Four Natures of TCM.2. Based on Biothermodynamic study and combined with Fingerprint Chromatography and computer pattern-identification, the relativities between biothermoeffect and chemical composition were synthetically analyzed. The study showed that the different preparation made contents of al, Jatrorrhizine, Epiberberine and Coptisine alteration and then natures transformation; the different compounds madecontents of al, Coptisine, Evodiamine and Berberine alteration especially Evodiamine change greatly and then natures transformation.The researches showed that the evaluation and screening system based on Thermodynamics and combination of Thermodynamics and Fingerprint Chromatography respectively could not only concentrate on the Natures and active essences but also elevate the Natures and pharmacological effect of TCM. Owing to the characteristics of rapid speed, microamount, high effective and widespread, this system could provide new method for Four Natures evaluation, treatment principle, preparation, compatibility, quality evaluation, effective substances screening and new drug research and development, etc. |
