Objective To compare the effects of immunological tolerance induced by administration of allogeneic splenocytes through portal vein(portal venous tolerance) and intestinal tract(oral tolerance) .Methods Administration of the splenocytes of the SD rat to the Wistar rat, through portal vein or intestinal tract .Allogeneic skin transplantation was established a week later .The survival time of the grafts was observed and the immunological states of the recipients were evaluated after skin transplantation. Result The graft mean survival time(MST) in the portal venous tolerance group,the oral tolerance group were 15.8±2.1d, 13.0±1.3d respectively,significantly longer than that in the control group(P<0.01); MST in the portal venous tolerance group was significantly longer than that in the oral tolerance (P<0.01). Measurement of CD25+ T lymphocytes and T lymphocytes subsets showed low level of transplantation reaction after administration of the allogeneic splenocytes. Conclusion The effects of immunological tolerance induced by administration of allogeneic splenocytes through portal vein was better than that of through intestinal tract. ...
