Colorectal carcinoma is one of the most common cancers with high mortality. Invasion and metastasis of colorectal carcinoma is the most direct cause to death. In recent years, with the development of cell and molecular biological skill, some effective methods have been used in studying the mechanism of invasion, metastasis of colorectal carcinoma.The NF-κ B transcription factors regulate the expression of many genes, The role of NF-κ B in immunological response, inflammation, cell survival and cell proliferation, particularly in carcinogenesis and tumor cell invision, metastasis has been studied extensively in the past decade. Recent studies have indicated that NF- κ B was significantly high in tumor cell strain and in tumor tissue, but there are Altercations in tumor celldifferentiation, metastasis, and cilinic staging of tumor.With immunhistochemical staining, we examined the expressions of NF-κB p65 in colorectal carcinoma and adjacent normal epithelial cells. By comparison of different expressions in different lesions this paper evaluates the correlation of NF- κ B expression in colorectal carcionma. Materical and methods:65 samples of colorectal carcinoma, 42 cases of adjacent normalcolorectal mucosa were taken into this study. These samples were odtained from patients operated in Colorectal Surgical Department of the First Affiliated Hospital of Zhejiang University from March 2001 to November 2001,. NF?κB activation was determined by immunohistochemical analysis of formalin-fixed, paraffin embedded samples. We quantified cytoplasma staining of p65 as a marker of the positive cells, combining with the intensity, the semiquantitative. Statistical analysis was performed with the x2 test and exact test. Results and disussion:1. NF-κκ B P65 staining was significantly enhaced in the cytoplasm of colorectal carcinoma cells in comparison to that in adjacent normal epithelial cells.2. NF- κ B P65 was correlated to tumor histological grade. Expression of NF- κ B p65 significantly high in high grade than low grade.3. NF- κ B P65 was not correlated to the age, sex, cilinic staging, lymph node metastasis and tumor size.Conclusions:1. Expression of NF- κ B was sighnificantly high in tumor cells in comparison to that in adjacent normal epithelial cells. (p<0.01)2. Expression of NF- κ B was sighnificantly high in high tumor histological grade in comparison to that in low grade. (p<0.05)3. Expression of NF- κ B was not correlated to the age, gender, cilinic stage, lymph node metastasis and tumor size. (p>0. 05)4. Detection of NF- κ B can be a impersonal index to dignosis and therapy of colorectal carcinoma.
