The Repair Of Femoral Head Defects With BFGF/PDPB

Objective: Basic Fibroblast Growth Factor (bFGF) is a mitogen for many cell types, including osteoblasts and chondrocytes. It can stimulate angiogenesis and oateoblast gene expression. We developed a rabbit model that mimics the clinical situation with trapdoor bone-grafting. The objective of this study was to evaluate the effect of the repair of femoral head defect with bFGF/PDPB. Methods: Twenty four mature New Zealand rabbits were used in the experiment. After capsulotomy, a trapdoor was created in the anterolateral surface of the femeral head and a 5mm diameter defect was established. In the experiments, forty eight femoral head defects in twenty four rabbits were divided into three groups, which implanted with bFGF/PDPB, PDPB and nothing respectively. The rabbits were killed after injection through blood vessels with prepared Chinese ink in the 2nd, 4th, 8th weeks after operation, and then the femoral heads of each rabbit were taken off. The specimens of injection of Chinese ink were created. X-ray, histopathology and morphological changes of blood vessel were observed after the cessation light microscope and image analysis. Results: Moderate-to-excellent result was seen both radiographically and pathologically at two weeks, four weeks and eight weeks after operationsin the groups treated with PDPB and bFGF/PDPB. Whereas untreated defects did not heal completely. Conclusions: Basic fibroblastic growth factor (bFGF) has the potential effect in treating avascular necrosis of femoral head.
