Objective: To explore the protective effects of Ginsenoside Rgl on the Obstructed Kidney and its possible molecular mechanism. Methods: Complete unilateral ureteral obstruction (UUO) models were produced in rats by left ureteral ligation. Thirty rats were randomly divided into three equal groups of ten. Sham-operated group: closing the abdominal cavity without left ureteral ligation. Non-treatment group: closing the abdominal cavity after left ureteral ligation. G-Rg1 treatment group: closing the abdominal cavity after left ureteral ligation, and perfusing stomach with Ginsenoside Rg1(120mg/kg/day).All rats were killed on day 7, the left kidneys were harvested immediately and fixed routinely. The morphologic analysis was performed through optical microscope. The expressions of Bax, PCNA, TGF-β1 proteins were detected by the methods of immunohistochemistry. The results were quantitatively analyzed by Computer Image Analysis System. Results: The increased expression of Bax and PCNA proteins in the tubular and interstitial cells, and the increased expression of TGF-pl proteins in the renal interstitium and glomeruli of the obstructed kidneys were detected compared with the sham-operated group. Ginsenoside Rgl significantly attenuated the expression of Bax proteins (P<0.05) and strengthened the expression of PCNA proteins (P<0.05) in the tubular epithelial cells of the obstructed kidneys compared with non-treatment group, Ginsenoside Rgl also attenuated the expression of TGF-pl proteins in the renal interstitium and glomeruli of the obstructed kidneys compared with non-treatment group (P<0.05) Conclusion: Bax possiblytook part in the apoptosis of the epithelial cells of tubules, and TGF-β1 possibly played an important role in interstitial fibrosis and glomerulosclerosis in the obstructed kidneys. Ginsenoside Rgl attenuated the apoptosis of the epithelial cells of tubules, accelerated the proliferation of epithelial cells and attenuated interstitial fibrosis and glomerulosclerosis. Ginsenoside Rg1 possibly had an obviously protective effects on the Obstructed Kidney.
