BACKGROUND & AIM:Breast cancer is one of the femal malignant tumors.Now,the incidence of breast cancer has reached 1,300,000 all over the world.The incidence of breast cancer is obviously increasing in our country and has been the first of femal malignant tumors in some major cities.The death of breast cancer is one third of the incidence of breast cancer,and has risked the health of women.The therapymethods of breast cancer includes neoadjuvant chemotherapy,exairesis,endocrine therapy,targeted therapy and so on.Because some patients can not tolerate operation chem;the patients with negative ER and/or PR are not sentive to endocrine therapy;the patients are not sensitive to targeted therapy;in addition,chems and endocrine therapy has many side effects;so people start paying much attention to the traditional chinese medicine which can treat breast cancer. The panix is traditionary Chinese herbal medicine,including ginsenoside which is active component.20(R)-ginsenoside-Rg3 has the apparentest effect against cancer,which inhibits tumorous growth and aversion in many ways.To take place tumor is a complex course,which includes many protein factors and many molecular mechanism.The mechanism against tumor of 20(R)-ginsenoside-Rg3 is one of investigative hot spots of Chinese medicine against tumor.The aim of this study is to explore the effect of Rg3 on induction apopotosis of human MCF-7 breast cancer cell line through up-regulating FAS protein.MATERAIL & METHODS:The cell proliferation inhibition was measured by 3-[4,5-dimethylthiazo-2-yl]-2,5 diphenyl tetrazolium bromide (MTT) assay. Cell cycle and apoptosis rate were analyzed by flow cytometry.The FAS of MCF-7 cell is detected by immunocytochemistry.RESULTS:The inhibitory effect of Rg3 on MCF-7 cell is increasing with the increase of concentration. IC50 was 200μg/ml Rg3 after treatment for 48 hours.The rate of the S state cell and the rate of apoptosis both increase by the detect of flow cytometry.The FAS protein of MCF-7 cell increase by Rg3.Conclusion:1. Rg3 can restrain hyperplasia of MCF-7 cells;2. Rg3 can induct apoptosis and accommadate cellcycle of MCF-7 cells.3. Rg3 can up-regulate expression of Fas protein of MCF-7 cells.
