Ginsenoside Rh2 is a compound with antitumor activity, however,the molecular regulatory mechanism underlying the antitumor effect ofG-Rh2, especially its non-organic specific antitumor effects, has not beenfully revealed. In this regard, using gene microarray and two-dimensionalelectrophoresis technology, combining with bioinformics methods, weexplored the gene and protein expression profiles of G-Rh2-treatedHL-60 cell line on the transcripteome and proteome levels, andcharacterized the signal pathways and molecular interacting networks, toelucidate the molecular regulatory mechanism underlying antitumoractivity of G-Rh2.Gene microarray was used to obtain gene expression profile afterG-Rh2 treatment. The Pathway analysis of gene expression profile wasperformed by bioinformatics software GenMAPP. Analysis of keydifferently expressed molecule was conducted by western blotting inmultiple tumor cell lines. The neutralizing antibody of key differentlyexpressed molecule was utilized to further functional research.Two-dimensional electrophoresis technology was used to obtain proteinexpression profile after G-Rh2 treatment, mass spectrum analysiscombined with bioinformatics methods were used to get the informationof differently expressed molecules.Gene expression profile of HL-60 cells after G-Rh2 treatment showsthat 76 genes are markedly modulated, the most significantlyup-regulated gene is TNF-α. The altered gene expression profile is inconcert with the apoptosis of HL-60 cells on the whole. The up-regulationof TNF-αexpression is confirmed by western blotting. The neutralizingantibody of TNF-αcomfirms the up-regulation of TNF-αis involved inthe apoptosis effect of G-Rh2 on HL-60 cells. Two-dimensionalelectrophoresis shows the altered protein expression profile of HL-60cells after G-Rh2 treatment, a significant up-regulated molecule caficurtinis concerned to apoptosis. G-Rh2 can induce the overexpression of TNF-αin HL-60 cells andother tumor cell lines.; The TNF-αsigal transduction pathway is a mainpathway mediating the apoptosis of HL-60 cells induced by G-Rh2.
