| Objective: The aims of this study were to investigate the expression of c-Met protein, metallothionein and ki67 antigen in colorectal cancer and their relationship with clinicopathological factors.Methods: 40 fresh samples were obtained from patients who underwent colorectal cancer operation in the gastrointestinal department of surgery, the second hospital, HeBei Medical University. For each patient, 3 samples were obtained, which were respectively from the loci of cancer, 1CM from the loci and 10CM from the loci. And 10 samples obtained from patients who underwent colorectal operation for non-neoplastic disease were used as control group. The expression of c-Met, metallothionein and ki67 were studied by means of immunohistochemical assay (S-P method). The level of the expression was correlated with clinicopathological factors.Results: 1 The results of c-Met immunohistochemical expression in cancerous, paracancrous and non-neoplastic tissue: c-Met protein was mainly expressed on cancer cell membrane, the level of positive expression was higher in poorly developed tissue than in well developed tissue. Thepositive expression rate of c-Met protein in the loci of cancer, paracancer, 10CM from the loci and control group were 52.5%(21/40), 52.5%(21/40), 0.0%(0/40) and 0.0%(0/10) respectively. The positive expression rate of c-Met in the loci of cancer group and paracancer group were higher than that of 10CM from the loci and control group (p<0.05). There was no significant difference between the loci group and paracancer group 1CM from the loci(P>0.05). There was also no significant difference between the control group and the group 10CM from the loci of cancer(P>0.05). 2 The relationship between c-Met expression and clinicopathological factors: There was a significant difference of the positive expression between the group of Dukes C 84.6%(11/13), and the group of Dukes A,B 37.0%(10/27). (p<0.05). The same as between the group of poorly developed tissue and well developed, between the group with lymph node metastasis and the group without lymph node metastasis, between the group with serosa ivasion and the group without serosa invasion, between the adenocarcinoma group and mucous adenocarcinoma group. (p<0.05) There was no significant difference correlation with age, sex and the tumor size.(P>0.05). 3 The results of the metallothionein immunohistochemical expression in cancerous, paracancrous and non-neoplastic tissue: in non-neoplastic tissue metallothioein was mainly expressed in cytoplastic, the stain pattern were uniform. Incancerous tissue the stain pattern were patchy, in most cases the nuclear stain could be found. The positive expression rate of metallothionein in the loci of cancer, paracancer, 10CM from the loci and control group were 55.0%(22/40), 55.0%(22/40), 100.0%(40/40) and 100.0%(10/10) respectively.
