Clinical Trial And Experimental Study On Heat Blood Stasis Of Qi-fen Syndrome In Seasonal Febrile Diseases

Purpose: To investigate weather there is heat-blood stasis at the stage of Qi-fen syndrome in seasonal febrile diseases, and guide the clinical practice.Method and Result: A controlled clinical trial was conducted: 30 cases of Qi-fen syndrome were screened out. The levels of endothelin-1(ET-1), 6-keto-PGF1 (6-K), and thromboxane B2 (TXB2) were measured by radioimmunoassay (RIA), and the levels of D-dimer (D-D) were tested by enzyme-linked immunosorbent assay (ELISA). Results show that the levels of ET-1, 6-K, TXB2, D-D and the rate of T/K in observed groups are significantly higher than those in the control group. In the experimental study, 32 rabbits were divided into four groups randomly (Group A, Baihujiahuzhang Group; Group B, Baihu Group; Group C, Control Group; Group D, NS Group). Endotoxin was injected venously in Group A, B and C with the dose of 15ug/kg (evil-attacking), while NS was injected in Group D with the same dose as control. The levels of ET-1, 6-K and TXB2 are tested by the blood sample before evil-attacking and 1 hour, 3 hours, 24 hours after evil-attacking respectively. At the same time of evil-attacking, before 2 hours and after a half hour, 1 hour of that, Group A were injected intraperitoneally with Baihujiahuzhang decoction (with the dose of 8ml/kg), Group B were injected intraperitoneally with Baihu decoction (with the dose of 7ml/kg). Results show as follows. 1. Thechanges of ET-1: ET-1 in Group A has the tendency of increase first and decrease lately, but no statistic difference was found. ET-1 in Group B increases at the second time and at the third time (compared with Group A and D, P<0.01-0.05) and returns normal at the fourth time. ET-1 in Group C increases at the second, third and fourth time (compared with Group A and D, P<0.01-0.05), remains higher after 24 hours (compared with Group A, B and C, P<0.05). At the second time that in Group C up to the top; There are no obvious changes in the Group D; that in Group A and D shows no significant difference through the experiment. 2. The changes of TXB2: TXB2 in Group A shows the tendency of increase first and decrease lately, but no statistic difference was found. TXB2 in Group B and C increases largely at the second time, and remains higher at the third and fourth time (compared with Group A and D, P<0.01-0.05), though it drops slightly at the fourth time. There are no obvious changes in the Group D, and TXB2 in Group A and D shows no significant difference through the experiment.. 3. The changes of 6-K: 6-K in Group A shows the increasing tendency, but no statistic difference was found. 6-K in Group B up to the top at the third time (compared with Group A and D, P<0.05), and returns normal at the fourth time; 6-K in Group C up to the top at the third time, compared with Group A and D, P<0.01, and compared with Group B, P<0.05, 24 hours later returns normal; There are no obvious changes in that of Group D. 4. The changes of T/K: There are no difference between Group A and D through the experiment; The rate of T/K in Group B and C increases largely at the second time, compared with group A and D, P<0.01 respectively, and compared with the first time P<0.01 respectively; T/K in Group B and C remains higher at the third and fourth time though it declines slightly, Group B and C compared with Group A and D respectively, P<0.05, and compared with the first time respectively, P<0.05. 5. Pathological observation: There are obvious congestion in the lungs, livers, kidneys, spleen, stomach and bowels in the rabbits of Group B and C, and there are macular bleedingspots on the surface of lungs; there are no obvious pathological changes in the Group A and D. Microscopic observation: Pathological changes are found in the organs of Group C and B, such as cell degeneration, tissue congestion or bleeding; slightly changes are found in the Group A; while there are no obvious pathological changes in Group D.Conclusion: 1.
