The anti-inflammatory and immune action of Tongbi Capsule was studied in rats, mice and guinea pigs. Tongbi capsule (Tb) is a Chinese herbal complex prescription that is come from traditional Chinese medicine and long clinical experiences. Tb is manufactured into capsule by scientific technology and used in the treatment of rhinitis and sinusitis. In our experiment, we observed the theraputic effects of Tb on acute, chronic and immune inflammation in rats and mice by using several study models, and its mechanism is discussed preliminarily. The purpose is to provide a scientific basis for clinical application and further research.. Methods and results are as follows:1.Effect on allergic rhinitis induced by TDI in guinea pigs. After allergic rhinitis was induced by 10% TDI olive oil. Dose: NS10ml/kg, Tb700mg/kg, Xf400mg/kg, The drugs were administered daily for 15 days. On the 16th day, mucous membrane of nose was taken out and fixed for being observed and taken photoes by light microscope and electron microscope. At the same time, the spleen and thymus were removed and weighed. The results showed that Tb remarkedly inhibited AR induced by TDI in guinea pigs, the effect accompanied by thymic atrophy, while the weight of spleen of rats were not affected. 2.Effect on delayed type hypersensitivity induced by DNCB in mice. Dose: NS20ml/kg, Tb1500, 750, 375mg/kg, Aspirin5mg/kg. The drugs were administered daily for 8 days. On the 2nd day and 5th day, 7%DNCB acetone solution was injected into dorsalis subcutis of mice. And 1%DNCB acetone solution was applied to the right earflaps of mice at 15th hours before last administered and DTH was induced. Weight difference between the same size of the two earflaps was indexed and degree of swelling and inhibition rates were calculated. The results showed that Tb significantly inhibited, in a dose-dependent manner, DTH induced by DNCB in mice. 3.Effect on the phagocytic function of the mononuclear phagocyte system in mice. Dose: NS20ml/kg, Tb1500mg/kg, Ind10mg/kg. After administrationof the drugs, charcoal particles was injected via the caudal vein, the blood was extracted from the postocular venous plexus at different times. Absorbance was measured with spectrophotometer. The clearance rate was calculated. The results showed that Tb significantly increased the phagocytic function of the mononuclear phagocyte system in mice.4.Effect on swelling of the earflaps of mice induced by xylene, croton oil. Dose: NS:20ml/kg, Tb1500,750,375mg/kg, Indomethacin 10mg/kg. After administration of the drugs, xylene or croton oil was applied to the right earflaps and inflammation was induced. Weight difference between the same size of the two earflaps was indexed and degree of swelling and inhibition rates were calculated. The results showed that Tb significantly inhibited, in a dose-dependent manner, the swelling induced by xylene and croton oil in the ear of mice.5.Effect on scald edema in rat. Dose: NS10ml/kg, Tb900mg/kg, Ind 10mg/kg. After administration of the drugs, scald edema was induced by immersing the right hind paws into hot water. Each volume of the right hind paws was measured at different time and inhibition rates were calculated. The results showed that Tb markedly antagonized scald edema of rats. 6.Effect on swelling of hind paws induced by injection of carrageenin in rats. Dose: NS10ml/kg, Tb900mg/kg, Ind 10mg/kg. After administration of the drugs, carrageenin was injected under the aponeurosis of the right hind plantar paws to induce inflammation. The approaches to the measurement and calculation are the same as 2. The results showed that Tb dramatically inhibited the swelling of hind paws induced by carrageenin and the effective time lasted for 6 hours.7.Effect on proliferation of granuloma induced by cotton-pellets in mice. Two aseptic cotton-pellets were implanted in both subcutameous scapular regions. Dose: NS20ml/kg, Tb1500, 750, 375mg/kg, Tbv5000mg/kg. The drugs were administered daily for 7 days.
