Objective To investigate the expression of apoptosis inhibition gene, survivin and its relationship with the clinical-pathological features, and with the proteins of bcl-2, and bax in uterine endometrial carcinomas. Methods A total of 102 uterine endometrial tissues samples were used in this study, including 56 samples of carcinoma tissues, 28 samples of hyperplasia endometriums and 18 samples of normal endometrium tissues .The survivin mRNA expression in different sample were detected by semi-quantitive reverse transcription-polymerase chain reaction(RT-PCR), glyceraldehyde-3- -phosphate dehydrogenase(GAPDH) was used as internal control. The detected ratio of survivin mRNA to GAPDH mRNA by compute image analysis system represents the value of survivin mRNA expression in every sample. The values were evaluated for associations with menstrucation, surgical stage, histoligical grade, statistical analysis was performed by using SPSS11.0 statistics software. Using streptavidin -biotin peroxidase (SP) method of the immunohistochemistry, We also examined the expression of the proteins survivin, bcl~2 and bax in normal uterine endometrium tissues and uterine endometrial carcinoma tissues.Results 1.Survivin mRNA were detected in 3/18(16.7%) normal endometriums, 11/28(39.3%) hyperplasia endometriums and 46/56(82.1%) endometrial carcinoma tissues, the expression of survivin mRNA in three group above has significant diffrence (P=0.0122, x2=8. 808) .2. Among endometrial carcinoma group, The expression level of survivin mRNA in low-stage and high-grade groups were sinificantly lower than inhigh-stage and grade groups (P=O. 00384 and 0. 00273). The expression level of survivin mRNA has no association with menstrcution statue and age (P= 0. 840, and 0.9596 ).3.The protein of survivin was expressed in 73.2% (41/56) cases of endometrial carcinoma tissues, 32. l%(9/28)cases of hyperplasia endometriums, however, not expressed in normal uerine edmetriums. The expression of survivin was related to the clinical-surgical stage and the tumor grade (P=0. 02011, 0.00095; x2=7.813, 13.911), and was strongly associated with the expression of bcl-2, bax (P=0. 01969, <0.001; x2=5.4389, 15.789) .Conclusions 1. Survivin gene reexpressed and overexpressed in endometrial carcinoma tissues.2. The expression of survivin in carcinoma tissues has sinificantly association with surgical stage, hitological grade.3. Survivin could play an important role in early carcinogenesis of endometrium, and is a signnif icantly potential prognostic parameter of worse outcome in uterine endometrial cancer patients.4.The protein of survivin has relative associated with the expression of the protein of bax and bcl-2.
