PrefaceThe frequency of humen breast tumor in women is very high. Fi-broadenoma is the most common benign, and the breast cancer is the majority of malignant tumor. The breast cancer do great harm to female. At present, operation treatment and subsequent adjuvant other treatment is the treatment of choice. Relatively speaking, the survival of management of early breast cancer is better. But because the etiology of breast cancer is unclear, it is difficult to suggest exact etiological prophylaxia. Augmentation of study in this aspect have a important real significance. There has increasing interest in the related (3 - catenin and its complex.b - catenin is a 92 -95KD cytoplasm protein, which can interact with multiple protein to perform multiple biological function. Firstly, b - catenin is the downstream factor of Wnt signaling pathway. Wnt signaling give rise to increasing of b - catenin levels in the cytoplasm, when the latter reach a critical amount, it can translocate to the nucleus where it serve as a transcription factor to activate target gene, regu-lating differentiation and proliferation of cell. b- catenin is the critical regulation point of Wnt signaling pathway, so its role in the tumor occurrence has been followed with great interest. In addition, b -catenin is main cytoplasmic component of E - cadherin/catenin complex, which play a key role maintenance of polarity and integrity of epithelia. The abnormal of b- catenin or/with E - cadherin, which result in abnormal in architecture and funcation of the whole complex, play a role in tumor invasion and metastasis. Several studies have demonstrated an association between expression of b - catenin and E - cadherin/catenin complex and breast cancer occurrence , invasion and metastasis. But there is few study about the relationship between breast intermediate - high grade atypical hyperplasia and expression of b - catenin and E - cadherin; and there is still a argument about relationship between expression of E - cadherin and breast cancer with lymph nodes metastasis. In view of the above, we have examined the expression of b - catenin and E - cadherin in primary breast tumor to investigate its significance and mechanism by super-sensitive immu-nohistochemisty S - P method.Material and MethodsFormalin - fixed, paraffin - embedded breast tumor tissue specimens were retrieved from the archival material of the Department of Pathology, No. 2 clinical Hospital affiliated China Medical University. Specimens were obtained from 87 female patients operated for primary breast tumor. They were divided into four groups according to pathological diagnose; Normal breast tissue around the breast case; breast intermediate - high grade atypical hyperplasia; breast cancer withoutlymph nodes metastasis; breaist cancer with lymph nodes metastasis; and they were marked as A , B , C and D, respectively. The monoclonal antibody to b - catenin , the monoclonal antibody to E - cad-herin and S - P kit were purchased from ZhongShan Company, Beijing. The other secondary reagents were provided from department of PathologyFour - micrometre thick sections were mounted on coated slides, two pieces for each specimens, deparaffinize in xylene, rehydrated in graded alcohols and incubated with 3% H2O2 for 10min to block endodenous peroxidase activity. After antigen retrieval, normal serum was applied for 30min to block nonspecific antibody binding. Subsequently, sections were incubated at 371 with the primary antibodies for 0. 5hours, and the secondary antibodies and S - P kit at 37 C for 0.5hours. Phosphate -buffered saline (PBS) was used in all washing steps, 5min+ 3times. Finally, diaminobenzidine was used as the chromogen and the sections were lightly counterstained with Harris haematoxylin; dehydrated; hyalined; coverslipped. As a negative control, the primary antibody was replaced by PBS.
