| ObjectiveThe entity that gastrointestinal lymphoma arising from mucosa -associated lyphoid tissue ( MALT) , was first described in 1983 by Isaacson, an English doctor of pathology. From then on, people pay more and more attention to this kind of lymphoma. Before, the lym-phoproliferation of gastric mucosa with proliferation of lymphoid follicle was called pseudolymphoma". With research going on, the relationship between cytogenetics abnormalities and pathological diagnosis of tumor was noticed recently. Trisomy 3 plays an important role in cytogenetics of gastric mucosa - associated lyphoid tissue lymphomas. At present, the pathogenesis of gastric lymphoma is not clear. The roles of HP and trisomy 3 in gastric lymphomas are not defined.While this kind of internal research is very limited since the incidence of gastric lymphomas is rather low, about 1%-4% of malignant gastrointestinal tumor. In most districts, there is only 2-3 cases every year. Therefore, it is impossible to collect many fresh samples in short time. While conventional karyotype analysis demand meta-phase cells by cell culturing of fresh tissue, though it can investigate chromosome abnormalities of neoplasm cell. That approach cant be applied in reviewing research and is limited in routine sections. Basedon all of above, using flurescence in situ hybridization (FISH) on intact formalin - fixed, paraffin - embedded tissue may be useful in resolving this problem.Our research applied fluorescence in situ hybridization ( FISH) and immunohistochemistry on paraffin samples to detect the pathohis-tological and genetic characters of 20 cases of gastric lymphomas. Which provide some evidence for the relation among trisomy 3 N infection of HP and gastric lymphomas, meantime discuss it. At the same time, this research may give some experience of doing FISH on paraffin samples with internal condition.Material and methods1. material: 20 cases of gastric lymphomas come from the first affiliated hospital of China Medical University and Liaoning Tumor Hospital ( 1979-2002). The age of patients including 10 male and 10 female is 19-72 with average of 53.4. The tissue samples were fixed by formalin and paraffined, cut into 4m sections and stained with routine HE.2. The cases were stained by immunohistochemistry (rabbit and HP, 1:20,DAKO) with SP method.3. FISH; Briefly, several 40 - m sections were deparaffinized in xylene, washed in 100% ethanol, and resuspended in protease K, then homogenate, filtrating, centrifuge, smearing. Nucleus on slides were fixed by 4% polyformalin. Probe and nucleus were denatured at 90 C for 15 minutes, followed by cooling with gradient ethanol in -20 C and prehybridization at 42 C for 2 hours, then incubation at 31 C for 14.5 hours. ( We design primer according to probe of D3Z1,and synthesized the primer in Shengong Company in Shanghai, marked it by ourselves). Slides were counterstained with PI and examined under oil immersion. In order to eliminate the error caused by the nucleus close together, we use separate nucleus and show complete form of nucleus with counterstained by PI. The signals marked by FITC are yellow - green and the nucleus are red.Results1. In the 20 cases of gastric lymphomas, 10 cases are MALT lym-phomas,9 cases are diffuse large B - cell lymphomas( DLBL) and 1 case mantle cell lymphoma( MCL)2. Trisomy 3: 8 of 20 cases (40% ) revealed trisomy 3. four of these cases are MALT lymphomas and the other four cases are DLBL.3. HP: 17 cases of 20(85% ) are positive of HP. The coherence between HP and trisomy 3 is 100%.Conclusions1. The rate of presence of trisomy 3 in 20 cases of gastric lymphomas is 40% , 40% in MALT lymphomas and 50% in DLBL. The similar rate between them suggests that trisomy 3 may play an important role along with evolution and transformation.2. In 17 (85% ) of 20 cases of gastric lymphomas, evidence of HP infection is absent. This supports the close relationship between gastric lymphomas and HP.3. We found that HP are all p...
