Pharmacodynamic Studies Of Kanggubisu On Experimental Osteoporosis

Kanggubisu(Kgbs) is a traditional Chinese prescription. It possesses "heat-clearing and dampness drying"function and is used for arthralgia due to damp-heat pathogen, pruritus due to damp pathogen, leaborrhagia due to noxious dampness,et al in clinical treatment. In order to investigate the efficacy of Kgbs prescription and its extracts (Kgbs I, II, III) on osteoporosis, the experimental models of osteoporosis (OP) induced by retinoic acid (RA), ovariectomy (OVX) and glucocorticoid (GC) were used in this study in rats. The bone mineral density (BMD), bone biomechanics and bone histomorphometry and the biochemical index of serum were measured. Moreover, the effects of Kgbs I, II and III on anti-inflammation, analgesia, and Kidney Yang Deficiency (KYD) mice were studied.In RA-induced OP, at the higher dose (po.28 days), kgbs I, II and III increased the BMD of femur. Kgbs II (50-100 nig/kg) and III(20-80 mg/kg) increased the bone mineral consistency (BMC) offemur. Kgbs III showed remarkably improving effect on morphological construction in rat femur. These results suggest that Kgbs has significantly preventable and therapeutic effects on OP induced by RA in rats.Kgbs I (0.75-1.5 g/kg, po,90 days) and III(20-80 mg/kg) raised the HMD in OVX rat. Kgbs I (1.5 g/kg) and II (25-50 mg/kg) and III (40-80 mg/kg) significantly increased the BMC of femur in OVX rats. Kgbsl, II and III showed markedly improving effect on morphological construction in femur of OVX rats. These results suggest that Kgbs has significantly preventable and therapeutic effects on OP induced by OVX in rats.Compared with the model group, kgbs II (50-100 mg/kg, po 90 days), III (80 mg/kg) increased the HMD in GC-induced OP in rats. Kgbs I had slighter effects in raising the HMD. Kgbs I (0.75-3 g/kg), II (100 mg/kg) and III (80 mg/kg) increased the BMC to the levels almost similar to that of the saline group. Kgbsl, II and III partly improved morphological construction in the femur of rats with OP induced by GC. These results suggest that Kgbs has partly preventing and therapeutic effects on OP induced by GC in rats.Kgbs I, II and III all could antagonize the changes of body tempreature, locomotor activity of KYD mice induced by GC. Kgbs I,II and III, at the all dose, antagonized the atrophy of thymus in KYD mice.Kgbs I, II and III had anti-inflammatory activities. Kgbs I (1.5-6 g/kg, po 7 days), Kgbs II (50-100 mg/kg) and Kgbs III (40-160 mg/kg) had significantly suppressing actions on ear edema induced by xylene and the increase in vascular permeability induced by acetic acid in mice. Three doses of Kgbs I, II and III also markedly abated the proliferation of granuloma induced by cotton-pellet in mice.In analgesic experiments, it has been proved that kgbs I, II andIII have analgesic actions. Kgbs I (3-6 g/kg, po 7 days), Kgbs II (50-100 mg/kg) and Kgbs III (40-80 mg/kg) could decrease the number of writhing induced by acetic acid in mice. Kgbs I (1.5-3 g/kg) increased the pain threshold in the hot-plate test in mice.These results indicated that kgbs I, II and III have obviously protective effects on OP in experimental animals. The results provided the pharmacological evidence for Kgbs in the treatment of OP.
