Objective: The aim of the study was to explore the correlation between HLA-DR allelic polymorphism and hereditary susceptibility of nasopharyngeal carcinoma in the Han race in Hunan province. Methods: 93 NPC patients and 93 normal individuals were genotyped for HLA-DR locus by PCR/SSO technique. We amplified HLA-DR gene by applying the primers provided by the 11th International Histocompatibility Workshop and Conference. The products of amplification were hybridized with sequence specific oligonucleotide probes tagged with [γ-32P] ATP. After analyzing the hybridization patterns, the samples were divided into groups. Then we amplified HLA-DR gene by group-specific primers. The products of group-specific amplification were hybridized with SSOP. We assigned HLA-DR alleles of every sample by analyzing hybridization patterns. The frequencies of alleles, haplotypes and genotypes of HLA-DRB1, -DRB3, -DRB4, -DRB5 in NPC group were compared with those in normal group by chi-square test.Results: The frequency of HLA-DRB1*0801 in NPC group was much higher than that in normal group(x2=4.221, P=0.04, RR=2.02), the frequency of HLA-DRB1*1101 was significantly lower in the former than that in the latter(x2=5.284, P=0.022, RR=0.32). There was no significant difference after Bonferroni correction (Pc>0.05). Conclusion: The results indicate that there is no significant association between nasopharyngeal carcinoma and HLA-DR locus in the Han race in Hunan province.
