ObjectiveTo investigate the genetic polymorphism of D11S4463and D17S974locus in Han population and to prepare their standard allelic ladders using molecular cloning technique.MethodsPolymorphism of the D11S4463and D17S974locus in600unrelated individuals was analyzed by fluorescence PCR and ABI3130XL Genetic Analyzer. Molecular cloning technique was employed to construct standard allelic ladders.ResultsD11S4463and D17S974locus showed high polymorphism in the Hans. Nine alleles and eight alleles were found respectively. The heterozygosity were0.735and0.747, the matching probability were0.089and0.115, the discrimination power were0.911and0.885, the polymorphic information content were0.73and0.70, the probability of paternity exclusion were0.485and0.505, respectively. Standard allelic ladders were constructed by molecular cloning successfully.ConclusionThe D11S4463and D17S974locus showed high polymorphism and pronounced robustness for genetic research and forensic application. Standard allelic ladders constructed by molecular cloning technique are useful in forensic science.
