Leukaemia is a malignant hematologic disease which threaten health of human being very seriously. While killing tumor cells traditional radiotherapy or chemotherapy will damage normal tissues and cells, especially hematopoietic tissue and cause hematopoietic disfunction. So induction of leukaemia cell differentiation will become a new method for the treatment of tumor. To investigate the effect of some cytokines and chemical drugs to the growth and differentiation of leukaemia cells, human myeloid leukaemia HL-60 cells as the target cell was used, and the results was obtained by cell proliferation assay, cell cycle analysis, in situ hybridization, immunohistochemistry , NBT reduction assay. The effect of rh-LIF used in single or combined with Ara-c on the growth and differentiation of HL-60 cell line and it's possible mechanism were observed. The results indicated as follows:HL-60 cell line could differentiate into monocyte and macrophage cell line in the presence of rh-LIF and differentiate into monocyte cell line in1. low dosage of Ara-c.When both of them was used simultaneously, the effect of differeniation induced by rh-LIF was weakened by Ara-c.2. Inhibition on HL-60 cell proliferation of rh-LIF appeared to be dosage dependent, effect of rh-LIF in 1u-1000u/ml was exhibited and obvious in 10000u/ml (p<0.05).3. Cell cycle analysis indicated that about 50% of HL-60 cells stagnated at G0/G1 stage in the presence of rh-LIF or low dosage Ara-c.4. rh-LIF and low dosage Ara-c increased the expression of P53 mRNA, P53 protein and p21 protein(p<0.05), and the effect of former was stronger than the latter. P53 expression induced by rh-LIF was time dependent.5. rh-LIF and low dosage Ara-c inhibited the expression of oncogene bcl-2(i.e apoptosis inhibiting gene ) in HL-60 cell line and the effect was time dependent.
