Objective:To improve theory of pathogenesis and clinical therapy, investigation of immune regulation status of the patients with different courses of condyloma acuminate (CA) was performed. The numbers of natural killer T (NKT) cells weredetected, and tkc Cession of CD 154 On T OQllfi OF OytOltiM IL4 2nd IFN=V OR bOth T cells and NKT cells from the patients with condyloma acuminate (CA) in response to in vitro stimulation were determined and compared with that of the normal.Methods:T cells and NKT cells were cultured in whole blood. The cellular surface molecules TCRVcc24 on NKT cells, CD 154 or intracellular cytokine IL-4 and IFN-Y expressed by both CDS T cells and NKT cells in response to in vitro stimulation were directly labeled by immunofluorescence. The mononuclear cells were separated by lysing the red blood cells and detected by flow cytometer.Results:1 The ratios of NKT cells to CD3+T cells from human peripheral blood of the short-history CA (SH-CA) group and long-history CA (LH-CA) group were (0.94 + 0.28)% and (0.77 + 0.21)% respectively, both significantly lower than that of the normal group (1.34 ±0.42)% (P<0.05).2 The expression rates of CD 154 on CD3+T cells of SH-CA group and LH-CA group in response to in vitro stimulation were (42.32±5.51) % and (38.72±4.96)% respectively, both significantly lower than that of the normal group (54.65±3.01)% (P<0.05). In the presence of monensin, the expression rates ofIL-4 on CD3+T cells of SH-CA group and LH-CA group in response to in vitro stimulation were (2.77 + 0.54) % and (2.81+0.43)% respectively, both significantly higher than that of the normal group (1.44 + 0.34)% (P<0.05), while the expression rates of IFN-y of LH-CA group were (32.18 + 6.78) %, which was significantly lower than that of the normal group(45.48 + 4.09) % or the SH-CA group (47.02+4.03) % (P<0.05).3 The expression rates of CD69 on NKT cells of SH-CA group, LH-CA group and normal group in response to in vitro stimulation were (99.35 + 0.61) %, (97.45 ±0.45)% and ( 94.69 ±2.58)% respectively, without significant difference between them(P>0.05). In the presence of monensin, the expression rates of IL-4 on NKT cells of three groups in response to in vitro stimulation were (48.62±5.44) %, (46.64±3.45)% and (50.53±4.55)% respectively, without significant difference between them(P>0.05), while the expression rates of IFN-y on NKT cells of SH-CA group and LH-CA group were (35.19 + 6.98)% and (33.28 ±5.34)% respectively, both significantly lower than that of the normal group (49.72 ±5.38)% (P<0.05).Conclusions:The level of CD 154 expressed by T cells from human peripheral blood of patients with CA in response to in vitro stimulation is lower than the T cells from normal persons. The amount of NKT cells from the peripheral blood of CA patients decreased, and the expression rates of IL-4 or IFN-y on both T cells and NKT cells in response to in vitro stimulation from CA patients were both abnormal, implicating the deficiency of immune regulation of the patients with different courses of CA. These data would provide some guidance for the immune therapy of CA.
