Objectives: To investigate the role of CD40 signaling in the pathogenesis of condyloma acuminatum and to provide new ideas for the clinical treatment of condyloma acuminatum.Methods:The warts of 12 patients with condyloma acuminatum were collected,and12 normal foreskin tissues were collected as control.The expression level and distribution of CD40/CD154 in warts and normal foreskin tissues were detected by experimental methods such as Real-time PCR,immunohistochemistry,and immunoblotting.The expression level of IL-12 in warts and normal foreskin tissues was detected by Real-time PCR method.Finally,the differences of the expression levels of CD40/CD154 and IL-12 in warts and foreskin tissues of the control group were analyzed by statistical methods.Results: 1)By using Real-time PCR detection technology,we found the expression levels of CD40/CD154 and IL-12 m RNA in warts of patients with condyloma acuminatum were significantly lower than those in the foreskin tissues of the control group(P <0.05).2)Immunohistochemistry showed that the expression levels of CD40 and CD154 in the epidermis and dermis of warts of patients with condyloma acuminatum were lower than that in the foreskin tissues of the control group.3)The results of immunoblotting showed that the expression levels of CD40 and CD154 in warts of patients with condyloma acuminatum were lower than that of the foreskin tissues of the control group,and the difference was statistically significant.The results were consistent with the results of Real-time PCR and immunohistochemistry.Conclusions: The expression levels of CD40/CD154 and IL-12 in warts of patients with condyloma acuminatum are significantly lower than normal foreskin tissues.CD40 signaling is involved in the pathogenesis of condyloma acuminatum.
