| Part I Literature Review and StudyWe studied the knowledges of Modem Medicine and Traditional Chinese Medicine(TCM) to lung cancer.Then we discussed the basic and significance of the research.Part II Clinical StudyObjectiverTo observe the therapeutic efficacy of Qining Injection(Sodium Cantharidate Injection) in treating medium or late stage non-small cell lung cancer(NSCLC). Methods.72 patients with stage III-IV NSCLC were divided randomly into three groups. In Group A(as a treated group),31 patients were treated with Qining Injection alone.In Group B(as a control group),20 patients were treated with chemotherapy alone.In Group C,21 patients were treated with chemotherapy combined with Qining Injection. The therapeutic efficancy of the three groups were evaluated after one course of treatment. ResultsiThe Group A had a better effect in the remission of symptoms such as cough,polypnea,debility than Group B(P<0.05 or P<0.01).There was no significant difference between Group A and Group B in the remission of symptoms such as hemoptysis,ignis,pectoralgia(P>0.05).The complete and partial remission rate(CR+PR) of Group A was lower than that of both Group B and Group C(P<0.05).The stable rate(CR+PR+NC) of Group A was higher than that of Group B and lower than that of Group C,but there was no significant difference between them(P>0.05).The stable rate of Group C was higher than that of Group B(P<0.05).Both Group A and Group C had a better effect of raising the quality of life and weight than Group B(P<0.05).Compared with Group B,the distant metastatic rate of both Group A and Group C was statistically lower(P<0.05).Qining Injection could increase the percentage of CD3+,CD4+ in T-lymphocyte subcategory statistically while the change of T-lymphocyte subcategory in Group B was not significant The percentage of CTV and CD4+/CDg+ in T-lymphocyte subcategory increased statistically in Group C.The Qining Injection had no severe adverse reaction wheras the chemotherapy had. The rate of bone marrow depression in Group C was lower statistically than that in Group B(P<0.05).Conclusion: Qining Injection could alleviate the symptoms,improve the quality of life,increase the weight,improve the immunity and stabilize the tumor in the medium or late stage NSCLC patients. Qining Injection could enhance the efficacy and reduce the toxicity of chemotherapy.Part III Experimental StudyObjective:To study the inhibition effect of Qining Injection on the proliferation of humanpulmonary carcinoma cell LAC and evaluate the influence of apoptosis and the expressionof p53 gene and bcl-2 gene.Methods:To investigate the effect of Qining Injection on the proliferation of humanpulmonary carcinoma cell LAC by Ml 1 methodTo test the apoptosis using FlowCvtometry(FCM) analysis.To examine the protein expression of the p53 gene and bcl-2gene using Flow Cytometry(FCM) analysis.Results:1. Qining Injection could suppress the growth of human pulmonary carcinoma cellLAC significantly in a obvious dose dependent manner. 2.After exposure of LAC to QiningInjection ,the cell cycle distribution was changed compared to control, the percentage ofcells in Go/Gi phase increased while it decreased significantly in S phase. Apoptosis ofpartial LAC cells was induced by Qining Injection and Sub- Gi peak was detected withFlow Cytometry.S.After exposure to Qining Injection,the p53 gene expression of LACincreased while the bcl-2 gene expression of LAC decreasedInclusion.Qining Injection could suppress the proliferation of human pulmonarycarcinoma cell LAC significantly. The apoptosis which induced by Qining Injection wasassociated with the changes of the protein expression of p53 gene and bcl-2 gene, possibly. |
