Objective:To observe the clinical efficacy of Shenqi Fuzheng injection combined with interferon-? in the treatment of advanced primary liver cancer and study the molecular biological mechanism of Shenqi Fuzheng injection combined with interferon-? on inhibiting the liver cell growth.Methods:The clinincal study:41cases of inpatients and outpatients with advanced primary liver cancer were selected and randomly divided into the study group which contained24cases of inpatients and outpatients with advanced primary liver cancer and the control group which contained21cases. The control group was only given the treatment of recombinant human interferon-?-1b injection; The control group was given the treatment of recombinant human interferon-?-1b injection combined with Shenqi Fuzheng injection.The clinical total effective rates and survival time of the two group were analysed after3mounths',6mounths'and12mounths'treatment; The Karnofsky grade? changment of weight and the poisonous side effect of the two group were analysed after treatment; The experimental study:we designed the control group?the interferon group which was treated only by recombinant human interferon-a-1b injection?the shenqi group which was treated only by Shenqi Fuzheng injection and the combined group which was treated only by Shenqi Fuzheng injection and recombinant human interferon-a-lb injection.Next,we used the experimental methods of MTT and Transwell to study inhabitable effect on the four groups'abilty of proliferation and Invasion of the liver cancer cell MHCC97-L.At the same time, we used the experimental methods of RC-PCR and Western Blot to study inhibitable effect on the expressional volume of STAT1mRNA and VEGFmRNA of the liver cancer cell MHCC97-L and its production of STAT1protein and VEGF protein.Results:The clinincal results:?The total effective rate of6mounths'.12mounths' and24mounths of the study group was higher than the total effective rate of the control group and the differences between the two groups were significant(P<0.05); The survival time of12mounths'.18mounths' and24mounths of the study group was longer than the survival time of the control group and the differences between the two groups were significant (P<0.05);?The KPS grade and weight grade of the study group was higher than the KPS grade and weight grade of the control group and the differences between the two groups were significant(P<0.05);?On the set of toxicity,the two groups of patients had no ECG changes. The descend of leucopenia and plaelet. gastrointestinal reaction, liver damange. the rate of alopecia of the study group was lower than the descend of leucopenia and plaelet. gastrointestinal reaction, liver damange. the rate of alopecia of the control group and the differences between the two groups were significant(P<0.05);The experimental results:?MTT Experienment:the inhibitable effect on the combined group' s abilty of proliferation of the liver cancer cell MHCC97-L was higer than the interferon group's and the differences between the two groups were very significant(P<0.001);?Transwell Experienment:the inhibitablel effect on the combined group's abilty of invasion of the liver cancer cell MHCC97-L was higer than the interferon group and the control group's and the differences between the two groups were very significant((P<0.01and P<0.05));?RT-PCR Experienment:After16hours'action,the the combined group's expressional volume of STAT1mRNA of the liver cancer cell MHCC97-L was higer than the interferon group and the control group's and the differences between the two groups were very significant (P<0.05);After24hours' action,the combined group's expressional volume of STAT1mRNA of the liver cancer cell MHCC97-L was higer than the interferon group and the control group's and the differences between the two groups were very significant(P<0.05).At the same time, After2hours'action and4hours' action,the combined group's expressional volume of VEGFmRNA of the liver cancer cell MHCC97-L was lower than the interferon group and the control group's and the differences between the two groups were very significant(P<0.05).In summary,the Shenqi Fuzheng injection combined with interferon-? can upregulate the expressional volume of STAT1mRNA and downregulate the expressional volume of VEGFmRNA than the only interferon-?' action.?Western Blot Experienment:After16hours' action,the the combined group's expressional volume of production of STAT1protein of the liver cancer cell MHCC97-L was higer than the interferon group and the control group's and the differences between the two groups were very significant(P<0.01).After4hours'action,the the combined group's expressional volume of production of VEGF protein of the liver cancer cell MHCC97-L was lower than the interferon group and the control group's and the differences between the two groups were significant(P<0.01).In summary,the Shenqi Fuzheng injection combined with interferon-? can upregulate the expressional volume of production of STAT1protein and downregulate the expressional volume of production of VEGF protein than the only interferon-?' action.Conclusion:The clinical total effective rates of Shenqi Fuzheng injection combined with interferon-? in the treatment of advanced primary liver cancer is better than the clinical efficacy of interferon-? in the treatment of advanced primary liver cancer.The Shenqi Fuzheng injection combined with interferon-? can prolonged the patiens'survival time,improve the patiens'quality of life,and relieve the patiens'toxicity caused by interferon-?.At the sime time, the combintion Shenqi Fuzheng injection and interferon-a can effectively suppress the abilty of proliferation and Invasion of the liver cancer cell MHCC97-L and upregulate the expressional volume of STAT1mRNA and protein of STAT1and downregulate the expressional volume of VEGFmRNA and VEGF protein so as to suppress the the liver cancer cell MHCC97-L.The experimental results is in accordance with the clinical results,and these may be the molecular biological mechanism of Shenqi Fuzheng injection combined with interferon-? on inhibiting the liver cell growth.
