Objective: To study the effect and mechanism of Chinese herbal compound Zhiganning on fatty livenTL) of rats induced by high fat diet and alcohol.Methods: 91 male SD rats were randomly divided into seven groups, i.e., large dose Zhiganning solution (group A), small dose zhiganning solution (group B), ursodeoxycholic acid (UDCA) solution (group C), lescol solution (group D), Qiangganxiaozhi mixture solution (group E), model group (group F), and normal control group (group G). The experimental period was 9 weeks. Except for normal control group, rats of six groups were fed with high fat diet (88% standard diet, 10% lard, 2% cholesterol) and 35% alcohol Iml/lOOg twice everyday. Prophylactic drugs were used similarly. All rats were sacrificed at 9th experimental weeks. The histological change in liver tissues were observed after Hematoxylin-eosin (HE) staining; The contents of triglyceride (TG) and cholesrerol (TC) in serum were determined by biochemical methods, as well as the contents of cholic acid and cholesterol in bile, malonyldialdehyde (MDA) and glutathione (GSH) in serum and liver. The activities of postheparinplasmalipoprotein lipase (LPL) and hepatic lipase (HL) were determined by colorimetric method.ResuIts:(l)The appearance of normal liver was crimson, smooth and fresh. The liver of rats in group F was significantly greater than that in group G, the color turned milk-yellow, the membrane was tensed and section was grease. Focal degeneration was observed. The liver of prophylactic groups were nearly normal in appearance, with crimson color, sharp-edge, smooth surface and section.(2)Fî–¯stological changes: The structure of hepatic lobules in normal rat was plain with normal sinusoids, cells arranged tidily and have no obvious pathological changes ,the structure of nucleus was plain. The liver of rat in group F lost normal arrangement and the structure of hepatic lobules disappeared, abundant cells of bullous steatosis were observed, diffusing from central veins to the edge. The volume of liver cells augmented and the cytoplast loosened, under-stain. The nuclear of bullous steatosis cell was pushed to one side by fat drops, and part of liver cells to be disintegration. The incidence of lipid degeneration above grade 3 was 55% while 90% above grade 2. The liver of prophylactic groups had marked improvement, the lipid degeneration level was significantly lower than that of group F (p<0.05 or p<0.01). Group A and E showed more powerful liptropic effect, without lipid degeneration above grade 3, less fat drops were observed, andthere was no obvious vacuolar or looseness in cells, some cells were mildly cloudyswelling. While the liver in group D were more effectively protected, the structure of hepatic lobules was plain, cells arranged tidily, but siousoids were mildly narrow and had more fat drops, the pathological change around central veins was slightly serious than that on edge, the structure of nucleus was plain.(3)The contents of triglyceride (TG) and cholesrerol (TC) in serum of group F were significantly higher than those of group G, while the TG and TC in group A were significantly lower than those of group F (p<0.05). The activity of postheparin-plasma hepatic lipase (HL) and total lipase activity in group B was significantly higher than that of group F (p<0.05), The activity of lipoprotein lipase (LPL) was higher than that of group F but had no significant difference between the two groups. Both of the lipase activities in group A were higher than that in group F but had no significant difference between the two groups. Compared between group A and B, the total activity had a trend of decrease while the LPL had a trend of increase, but there was no significant difference. Among the prophylactic groups, the activities in group A and B were significantly higher than that in group D but lower than that .in group E.(4)The contents of malonyldialdehyde (MDA) and glutathione (GSFf) in serum and liver of group F were markedly lowerthan that in group G.
